Objectives: To observe the influence of Danshen to the expressions of mRNA and protein of local hypoxia inducible factor-1α(HIF-1α) during acute ischemia reperfusion injury of skeletal muscles in SD rats, and its effects on the changes in erythrocyte aggregation index, erythrocyte deformation index, local swelling coefficient and histology of the left cremaster muscles of SD rats during ischemia reperfusion injury, therefore to discuss the influence of Danshen to the oxygen environment and hemorheology of skeletal muscle during ischemia reperfushion injury.
目的:通过建立SD大鼠左侧提睾肌的缺血再灌注损伤模型,观察丹参对SD大鼠骨骼肌缺血再灌注损伤局部缺氧诱导因子1α(Hypoxia Inducible Factor-1α,HIF-1α)mRNA及其蛋白表达的影响、对红细胞聚集和变形指数的变化、局部肿胀系数的变化以及组织形态学改变的作用,探讨丹参对缺血再灌注损伤骨骼肌局部氧环境及血液流变性的影响。

METHODS: A limb ischemia-reperfusion injury model in rats was established. The animals were randomly divided into three groups: control group, IR group and IPC group. The contents of diamide oxidase, nitric oxide, endothelin-1 (ET-1) and ratio of nitric oxide/endothelin-1 (NO/ET-1) in the plasma and the gut were measured. The leavels of myeloperoxidase, ratio of DNA chain, total nitric oxide synthase, inducible nitric oxide synthase and constitutive nitric oxide synthase in the gut were determined.

At the same time,the protective effect of mannital and VitC were assessed.Result:The serum level of creative phosphokinase and malon dialdehyde was higher,but superoxide dismutase activity was lower during reperfusion than in the ischemic period.The tissue structure and ultrastructure were damaged more seriously in reperfution period. After treated by mannital and VitC, the level of CPK and MDA in serum was lower, but SOD activity rasied, and the ultrastructure was better.
结果:再灌流组较缺血组血清CPK增高、MDA升高,再灌流组脑组织中MDA 升高,血清中SOD活力下降,再灌流组脑组织结构和超微结构破坏更加严重,应用自由基清除剂后血清中CPK降低,MDA含量降低,SOD活性升高,组织中MDA含量亦降低,超微结构较前好转。

Results showed that green firm callus and rooting were obtained with treatments of NAA 0.5-3.0mg/L, in which the NAA 0.5mg/L treatment appeared the optimal rooting result of 90%. White loose callus was obtained with treatment of 2,4-D0.1-3.0 mg/L.Callus can not be induced by adding BA solely at 0.5-3.0mg/L. There appeared bud redifferentiation only when appropriate concentration of combined BA,NAA and 2,4-D were applied.

Methods: Single lung in situ ischemic reperfusion animal model was used. 30 rabbits were randomly divided into three groups: sham group, ischemic/reperfusion group and ischemic preconditioning group. Wet to dry ratio and injured alveoli rate were determined. Ultramicroscopic structural change was observed under electron microscope. Heat shock protein-27 was detected with immunohistochemistry.
