The results indicated:0.1 mg/L 2,4-D or 0.1 mg/L 2,4-D+0.2 mg/L KT was optimal to induct the callus of carrot hypocotyls.The majority of the callus induced on the medium containing 0.1 mg/L 2,4-D regenerated embryoid,whereas,the callus induced on the medium containing 0.1 mg/L (2,4-D)+0.2 mg/L KT produced adventive buds.During the growth of regenerated plant,MS medium is available for rooting of seedling,and seedling regenerated on B5 medium must be placed on the B5 medium containing 0.1% IBA to develop root.

试验结果表明：0 1mg/L2，4 D或0 1mg/L2，4 D+0 2mg/LKT的激素组合有利于胡萝卜下胚轴的愈伤组织诱导；在0 1mg/L2，4 D的培养基上形成的愈伤组织主要以胚状体的形式再生，而在含0 1mg/L2，4 D+0 2mg/LKT激素组合的培养基上形成的愈伤组织主要以发生不定芽的方式再生；再生过程中，B5基本培养基上的苗子不易生根，需要附加0 1mg/L的IBA，而在MS培养基上苗子可直接形成根。

The plant expression vectors were constructed and two genes were transferred into A. thaliana and N. tabacum, antikanamycin plants were obtained.4. The trials of adventive bud regeneration directly and callus induction was conducted to select out the optimal explants and formula. The results revealed that double scale can produce 13~16 adventive buds per centimeter in MS medium added 10.0 mg/L 6-BA, 1.0 mg/ L NAA, 40 g/L sugar and 7.5 g/L agar. Scale, leaves, scape and different tissue of flower can be induced to produce callus in appropriate medium, but the filament with anther was the optimized explant.

构建了两个基因的pBI121和pCAMBIA1300植物表达载体，并通过农杆菌将上述基因转入烟草和拟南芥中，得到了卡那抗性的再生植株。4、以中国水仙不同部位外植体进行了不定芽直接分化和愈伤组织诱导分化试验，结果表明，双鳞片外植体在MS0+6-BA 10.0 mg/L+NAA 1.0 mg/L+蔗糖40 g/L+琼脂7.5 g/L培养基上可以大量地直接再生，再生芽数量平均可达13~16个/cm双鳞片，比传统的鳞茎盘之间分化提高30~50%；鳞片、叶片、花葶及花部不同组织在适宜的配方中均可不同程度地诱导出愈伤组织，但以带花药的花丝诱导出的愈伤组织最好。

We got the result: Bufo Raddei Strauch Tadpole (1) Bufo Raddei Strauch Tadpole was capable of lens regeneration, which originated from the epithelial cells at iris dorsal margin by dipigmentation.(2) Dipigmentation of Bufo Raddei Strauch by two methods: one was licked up by amoebocyte, the other one was flowed from the pigment cells.(3) In the lens regeneration cells of Bufo Raddei Strauch profiles of the mitochondria became complicated ribosome manifold, and the endoplasmic reticulum was cisternal dilation. During the process of deferentiation of lens fiber the cell nucleolus became little and chromatin shrinked.(4)The crystalline electrophoresis of Bufo Raddei Strauch Tadpole lens regeneration showed that during lens regeneration P-crystalline was expressed earliest then was γ-crystalline and α-crystallin, this result is consistent with newt.

结果表明：花背蟾蜍蝌蚪(1)具有晶状体再生的能力，其晶状体的再生来源于虹膜背缘的上皮细胞；(2)虹膜背缘去色素是转分化的前提，主要有两种方式：一种也是最主要的方式是在晶状体被摘除后虹膜外侧产生巨噬细胞，巨噬细胞吞噬虹膜色素上皮细胞中的色素颗粒；另一种方式是虹膜色素上皮细胞自身释放色素颗粒；(3)电镜观察发现虹膜色素上皮细胞转分化过程中线粒体有明显变化，核糖体增多，粗面内质网增多，晶状体纤维分化过程中细胞核逐渐凝聚；(4)SDS-PAGE电泳发现，花背蟾蜍蝌蚪晶状体再生中相关蛋白表达顺序与有尾类蝾螈相似，蛋白表达顺序依次是β-晶状体蛋白、γ-晶状体蛋白和α-晶状体蛋白，并且后两种蛋白几乎同时表达。
