The results showed that the Saccharopolyspora spinosa SP06081 protoplast yield was the highest under these conditions: the collected mycelium from SP06081 grown in Tryptic Soy Broth medium with 0.2% glycin for 48 h was treated by 0.1 mg/mL lysozyme at 28oC for 20 min, then plated on the R2YE medium with sucrose as osmotic stabilizer, the number of regeneration protoplast was up to 108/mL. The protoplast-regenerated strains exhibited changes in morphology and antibiotic production, 29.3% protoplast-regenerated strains was characterized by loose mycelium and abundant broken branches as did their parent. Among them, 58.2% strains presented the trend to positive variation in spinosad yield, with the highest spinosad yield of up to 582.0 mg/L, 85.6% higher than that of their parent.

Moreover, the thesis delves on the mechanical property, construction craft and quality control of the old asphalt mixture after recondition, how to apply the recycling mixture to the base, and cater to the needs of performance.

本文在国内外已有研究成果的基础上，根据国内普遍采用半刚性基层沥青面层这一结构特点，研究旧沥青路面的现场冷再生技术，提出了冷再生混合料的配合比设计方法，合理的基、面层掺配比例及水泥剂量，深入研究了旧沥青路面材料再生后的力学性能、施工工艺以及质量控制及检测，并将再生的混合料应用于路面基层，满足相应的路用性能要求。

Effects of gamma irradiation on shoot regeneration from leaf and bud growth in vitro of strawberry 1 The leaf was very sensitive to the gamma irradiation, and 'Toyonoka' was more sensitive than 'Akihime'. The LD50 (50% decrease of generation) of strawberry leaf was between 10 and 20 Gy. 2 Strawberry leaves were inoculated in vitro at 8 d-15 d after gamma rays irradiation, and the shoot regeneration rate from of 'Toyonoka' increased at 10 Gy and almost no difference from 20 Gy to 40 Gy. The positive effect dose for 'Akihime'.was from 10 Gy to 30 Gy, however, 40 Gy decreased shoot regeneration rate.

The results showed that NaN3 had great effects on TCL growth. Some TCL after treatment got white or browning and died and grew slowly, and the number of regenerated plantlets was reduced, and the regenerated plantlets became abnormal with leaves crimpled and thicker.
