The present studies showed that two cell populations were found in haemocytes: large cell with high granularity and small cell with low granularity by flow cytometry FCM on light scanttering pattern. Two distinct cell types were identified based on phase contrast microscope: one type of cell was dark and dioptric aberration, while the other was bright and dioptric strong. By Giemsa and H.E staining, cytoplasmic staining were heterogeneous and internal particles were obvious in one type of cell, while cytoplastic staining were homogeneous and internal particles were inexistent in the other type of cell. By transmission electron microscope, we found that the mitochondria, Golgi apparatus organelles were rich and internal particles were obvious in one type of cells, and contrary to the another cells.

流式细胞术光散射图谱显示血细胞被分两类，一类为颗粒度高的大细胞，另外一类为颗粒度低的小细胞；相差显微镜观察显示，血细胞可分为胞体暗、折光性差和胞体明亮、折光性强的两类； Giemsa和H.E染色显示细胞分为胞质染色不均一、胞内颗粒明显和胞质染色均一、胞内颗粒不明显的两类；透射电镜超薄切片观察显示，颗粒明显的细胞胞质内线粒体、高尔基体等细胞器较丰富，颗粒不明显的细胞胞质内细胞器较少；负染结果表明血细胞主要分为表面不光滑、突起明显和细胞表面光滑、突起较不明显的两类。

Results (1)ATP could inhibit the proliferation of U937 cells with inhibition rate over 43% after 48hour ATP treatment;(2)ATP treated U937 cells numbers increased obviously in G1 phase,and apoptosis peak appeared before G1 phase,apoptotic cell number was 3.4% for 24hour,and was 22.7% for 48hour of ATP treated U937 cells;(3) Nuclear chromatin condensed into sperical masses bound cytoplasma membrane formed apoptotic bodies which is shed from membrane surface into intercellular medium;(4) Apoptotic bodies were nigrosine staining negtive.

结果 (1)ATP对U937细胞的增殖有按摩明显的阻抑作用，加药48h后增殖的抑制率可达43%以上；(2)ATP处理的U937细胞周期发生改变，G1期细胞数按摩明显增多，ATP作用24h时G1期前出现亚二倍体峰——凋亡峰，凋亡细胞数为3.4%，作用48h时凋亡细胞数增多为22.7%；(3)ATP处理的U937细胞首先在核内染色质浓缩成半月形贴近核膜，逐渐向核膜外移动，进入胞浆内再移向质膜内外面，紧贴质膜外面再逐步脱离细胞体，进入细胞基质中，成为游离的凋亡小体。

Methods Cell culture,flow cytometry,HE staining,Nigrosine staining and electron microscopy were used. 根据起源于下地壳的中酸性岩浆岩的成分变化，可以约束其源区深度以及地壳最小厚度，为大陆板内成矿作用的深部过程研究提供重要信息。全球范围内，许多世界级斑岩铜矿和浅成热液矿化系统与同期的埃达克质岩存在密切的时空与成因联系，在国内主要成矿区带也识别出与金属成矿作用有关的埃达克岩。与俯冲过程无关的埃达克质岩的识别，使我们有可能结合其他地质证据构建完全不同于Sillitoe(1972)B型俯冲环境的斑岩铜矿成矿模式的大陆板内斑岩型金属矿床成矿模式。对于规模巨大、矿质主要源自地幔的热液矿床的形成，埃达克岩可能是必要条件，但不是充分条件。埃达克岩的成矿潜力通过地幔物质加入而获取，埃达克岩的成矿专属性由上地幔成矿元素分布特征决定。与俯冲有关的埃达克质岩浆之所以有利于成矿，重要的原因是存在大量来自俯冲板片的高压、高温流体以及俯冲板片熔融形成高氧逸度(fO2)的熔体，但产出在大陆板内背景、与俯冲无关的埃达克岩的成矿机制还不清楚。根据现代资源勘查理念，结合综合地质信息分析，埃达克质岩具有实际找矿意义。

We introduced improved primary mixed glial culture and different-attachment method to isolate and purify the OPCs, the cells were proliferated in serum-free medium, flow cytometry and immunohischemistry methods were employed to estimate the purity of cultured OPCs. Their abilities of differentiation and expression of trophic factors were identified by RT-PCR and immunostaining. Several methods including TUNEL and MTT were adopted to estimate the protective effects of conditioned culture medium from oligodendrocyte lineage cells on the primary cultured cerebellar granule neurons. Intravitreal transplant of OPCs, combined with retrograde fluorescent labeling the superior colliculus and intraorbital optic nerve transection, were used to investigate the protective effects of OPCs on the axotomized RGCs in vivo. Intravitreal transplantof OPCs or NSCs on the newborn rats, and retinal transplant of OPCs on the young rats were performed, to observe the myelin formation in the retina at different stages after cellular transplantation. Optic nerve transection was carried out on some rats with myelinated retinae, to study the influence of myelination on the injuried RGCs.

为此，本研究采用改良的胶质细胞混合培养与差速贴壁方法获得大鼠OPCs，使用无血清培养基进行扩增、培养，用免疫组织化学和流式细胞技术对培养细胞的纯度进行鉴定，对少突胶质系细胞表达部分营养因子的情况进行检测；采用TUNEL、MTT等方法对少突胶质系细胞条件培养基对原代培养小脑颗粒神经元的保护作用进行检测；将OPCs移植入成年SD大鼠玻璃体内，利用上丘逆行荧光标记技术，观察眼内移植的OPCs对眶内视神经切断时的视网膜神经节细胞的保护作用及其持续时间；将OPCs或NSCs移植入新生和幼年SD大鼠玻璃体或视网膜内，观察不同时期视网膜内髓鞘形成与分布特点，分析髓鞘的超微结构，并观察眼内髓鞘形成对损伤神经节细胞的保护作用。

二整体动物水平上探讨心肌有效转染途径：以PLacZ为报告基因，将成年大鼠分为空白对照纽(生理盐水心包腔内注射后超声导入)；转染质粒心包腔内注射组及其阴性对照组：质粒+微泡+酶的混合物心包腔内注射，超声导入；转染质粒舌下静脉注射组及其阴性对照组：质粒+微泡混合液经舌下静脉注射，超声导入。

Histologically, the seminiferous tubules contained numerous Sertoli cells and more Sertoli-spermatozoa complexes, accompanied by the depletion of Leydig cells with deeply stained nuclei. Mature spermatozoa were stored up in the epididymis, but only a few in the efferent ducts. In the second place was testicular atrophy（32/120; 26.7%）. The seminiferous tubules showed moderate to severe inactivity of spermiogenesis with evidence of only spermatogonia, spermatocytes and Sertoli cells. The Leydig cells were obviously decreased in numbers associated with decrease of lipid droplets in their cytoplasms. Testicular hypoplasia was the third disorders（22/120; 18.3%）. Only a few spermatogonia and Sertoli cells appeared without any spermiogenesis. The associated changes was decreased in Leydig cells and fibrous hyperplasia in the interstitium.Epididymal stones were sometimes found（12/120; 10%）. Grossly, yellowish-white nodules with various sizes and firm in consistency were observed in the epididymis and the front efferent ducts. Microscopically, the epididymal ducts were dilated with voluminous spermatozoa storage, even showed calcification in severe cases. The deposited calcium salts were stained positively by Von Kossa and Alizarin red methods.Amyloidosis was also detected in 10 roosters（8.3%）. Eosinophilic, homogeneous, amyloid-like substances were deposited mainly in the testicular interstitium and the periphery of blood vessels. These substances showed positive reaction by Congo red staining. Five roosters（4.2%）had Marek's lesions in the testis, epididymis and peripheral nerves with infiltration of pleomorphic lymphocytes. Only one case showed epithelial necrosis of seminiferous tubules accompanied by fibrous proliferation in the interstitium.

结果发现，在总共搜集的120个病例中，其中因年老所导致的产精力不佳为最多，占38例（31.7%），於镜下可见大量精虫黏附於Sertoli cell的表面，并可见Sertoli cell数量明显增多而Leydig cell明显减少，且其细胞核呈现浓染的现象，而在其副睪中仍可见到成熟精虫蓄留於管腔中，但在其输精管内却只有少量精虫存在；其次为睪丸萎缩，占32例（26.7%），镜下可见中度至重度无造精作用，其生精小管中只见到精母细胞、精原细胞及Sertoli cell存在，但Leydig cell数量明显减少且其细胞质内的脂质也明显减少；睪丸发育不全，占22例（18.3%），於生精小管内只见到精母细胞及少量Sertoli cell存在，不见造精细胞分化，於生精小管间质可见Leydig cell减少并伴随结缔组织增生；副睪结石，占12例（10%），肉眼下可在副睪及输精管前段见到黄白色大小不一的结节，触感坚硬，於镜下可见副睪管扩张并有大量成熟精虫蓄积，严重时可见钙化现象，以Von Kossa及茜素红染色均呈阳性反应；类淀粉沉著症，占10例（8.3%），镜下在睪丸间质及血管周围可见粉红均质样的物质沉积，以刚果红染色成阳性反应；马立克病，占5例（4.2%），镜下可在睪丸、副睪实质及周边神经内均可见到嗜碱性大小不一的淋巴样细胞浸润；睪丸坏死，占1例（0.8%），镜下可见生精小管上皮细胞坏死脱落及间质结缔组织增生。

The result using inhibitors of Ca〓 channels supported above observation. When Ca〓 channels in plasma membrane were blocked by LaCl〓, verapamil or nifedipine, and the Ca〓 channels in endomembranes opened naturally, extracellular CaM couldn't induce 〓 increase. When Ca〓 channels in endomembranes were blocked by ruthenium red and the Ca〓 channels in plasma membrane opened naturally, extracellular CaM induced Ca〓 increase in guard cells, but the extent of Ca〓 increase was lower than that of control.

使用质膜Ca〓通道抑制剂LaCl〓、异博定、尼群地平及细胞内Ca〓通道抑制剂钌红的结果支持这种推测：在质膜Ca〓通道被阻断、细胞内Ca〓通道仍正常开放的情况下，保卫细胞〓不能被CaM诱导增加；细胞内Ca〓通道被抑制、质膜Ca〓通道正常开放的条件下，细胞外CaM诱导保卫细胞内〓升高的幅度不能达到不使用Ca〓通道抑制剂的水平。
