Tetraploid embryos could be produced by electrofusion at the stage of two-cell embryos, which could develop to blastocysts followed by fusion of cytoplasm and nucleus and cleavage in vitro. During the fusion of cytoplasm, the DNA methylation levels of the fused embryos are as high as these of two-cell diploid embryos in vivo Then the embryos are rapidly demethylated when the nucleus begin to fuse, resulting in the lowest DNA methylation levels when the nucleus are fused completely. After that, the DNA methylation levels of the fused embryos are gradually increased until the morula stage. However, whereas an asymmetric distribution of DNA methylation is established in vivo-derived blastocysts with a higher methylation level in the inner cell mass than that in the trophectoderm, we can not detect the asymmetric distribution in most in vitro-derived tetraploid blastocysts.
结果表明:利用电融合方法制备的小鼠四倍体胚胎在体外培养体系中经历细胞质融合、细胞核融合及细胞继续分裂发育直到囊胚期的过程,在细胞质融合的时候胚胎卵裂球同体内体外培养二倍体胚胎一样,呈现高度甲基化状态;在细胞核开始融合的时候,甲基化水平急速下降,在细胞核完全融合的时候甲基化水平达到最低点;随着胚胎继续分裂,胚胎甲基化水平逐渐增加,在桑葚胚期甲基化水平最高;但是囊胚期四倍体胚胎内细胞团同滋养层细胞甲基化荧光信号没有差别,这与体内体外培养二倍体囊胚内细胞团细胞甲基化荧光强度高于滋养层细胞甲基化荧光强度不同。

METHODS: A total of 200 volunteers who need to receive spinal fusion in the Affiliated Hospital of Qingdao University Medical College were selected, including 102 cases received anterior cervical subcorpectomy combined with iliac bone implantation fusion following decompression, and 98 cases received posterior laminectomy for decompression combined with intertransverse process fusion. Meantime, 223 normal adults were served as the control group.

分析后路减压枕颈融合内固定术治疗合并寰枢关节脱位的寰椎枕骨化临床疗效。[方法]2005年12月至2007年6月间,对8例合并寰枢关节脱位的寰椎枕骨化患者在行颅骨牵引治疗一段时间(12~16 d,平均135 d)后采用枕骨大孔后缘扩大,寰椎后弓切除减压取自体髂骨枕颈融合Cervifix系统内固定术,手术后采用日本骨科学会神经功能评分分析临床疗效。[结果]8例患者随访6个月~2年,平均为15个月。8例患者无一例出现术后并发症,术前寰齿前间隙为5~9 mm,经颅骨牵引后为5~7 mm,寰枢关节脱位未能完全复位。手术前后JOA评分示神经症状均有不同程度恢复,影像学检查示枕颈区减压充分植骨区获得骨性融合,无一例出现内固定松动或断裂。[结论]合并寰枢关节脱位的寰椎枕骨化患者术前仔细评估影像学改变,采用颅骨牵引一段时间后行后路减压枕颈融合内固定术的治疗方案是合理可行的,且临床效果满意。

RESULTS AND CONCLUSLON: A total of 2 of 6 autogenous bone grafting cases suffered from graft bone dislocation, 1 suffered from pseudoarthrosis formation, and 1 suffered from donated ilium. Six of 21 autogenous bone graft with cervical plate-screw fixation suffered from loss of intervertebral height, and 3 suffered from pain of donated ilium.
