After hybridization of two DNA probes with target DNA, EDTA-〓 and β-diketone come close to each other, and an EDTA-〓-β-diketonate ternary complex having strong and long-lived fluorescence was formed, thus the target DNA was detected sensitively with a detection limit of 6 pM (0.6 fmol per assay) by time-resolved mode.

系统研究了使用EDTA-〓为标记物的石墨炉原子吸收光谱免疫分析法，并将之用于人血清中AFP和水中BSM的测定。该法中使用了EDTA-〓标记SA及生物素标记抗体或抗原，在96孔板中的免疫反应结束后，利用稀硝酸将反应产物分解，然后用石墨炉原子吸收光谱法对免疫反应产物进行测定。

Immunohistochemical method: The CD34 monoclonal antibody was selected as the first antibody, with the immunohistochemical SP procedure, then the pathological slice was displayed by DAB and stained again by hematein. One slice was used as a negative control and reacted with PBS fluid(0.01mmol/L) which was the substitute of anti- CD34.(the kits coming from Boster Biotechnology CO.LTD). The number of the vessels and vascular endothelial cells were displayed by immunohistochemical method.

免疫组化方法：病理组织切片以CD34单克隆抗体作为第一抗体，按照免疫组织化学SP步骤进行，DAB显色，苏木素复染，每组用0.01mmol/l的PBS液替代CD34抗体作阴性对照(试剂购自于武汉博士德公司)，应用免疫组化显示血管，对不同时期的血管生成情况及早期的炎性反映情况进行观察。

Compared with control,①the mean tumor weight of H22 of SXKA Granules three dose groups were decreased significantly(P.01,P.05),and the mean inhibition rates of SXKA Granules 20、10 g/kg dose groups were above 30%;②the mean tumor weight of S180 of SXKA Granules three dose groups were decreased significantly(P.01),and the mean inhibition rates of SXKA Granules 20、10 g/kg dose groups were above 32%;③the mean tumor weight of EAC of SXKA Granules three dose groups were decreased significantly ( P.01, P.05),and the mean inhibition rates of SXKA Granules 20 g/kg dose groups were above 38%;④the mean tumor weight of Lewis carcinoma of SXKA Granules three dose groups were decreased significantly(P.01, P.05),and the mean inhibition rates of SXKA Granules 20、10、5 g/kg dose groups were above 36%;⑤the mean tumor weight of W256 of SXKA Granules three dose groups were decreased significantly ( P.01, P.05),and the mean inhibition rates of SXKA Granules 20、10 g/kg dose groups were above 32%;⑵Compared with control,SXKA Granules 20、10 g/kg dose groups had extended the survial time of the P388-bearing mice respectively(P.01),and the mean prolong rates of SXKA Granules 20、10 g/kg dose groups were above 50%;⑶Compared with S180-bearing group, SXKA Granules 20、10 g/kg dose groups could increase the weight of thymus and spleen, Spleen index and thymus index were increased, SXKA Granules 5 g/kg dose group could increase thymus index(P.05);⑷As Compared with control group, SXKA Granules 20、10 g/kg dose groups could improve mouse serum half hemolysis value depressed by transplanted tumor dramatically(P.01), which revealed the SXKA granules could improve the mouse humoral immunity system;⑸SXKA Granules 20 g/kg dose group could increase of englobe indexαon S180-bearing mice remarkably(P.01), which indicated the SXKA Granules could improve their cellular immunity system.

对荷W256大鼠，生兴克癌冲剂20、10、5 g / kg三组的平均瘤重明显低于对照组(P.01，P.05)，生兴克癌冲剂20、10 g / kg组的平均肿瘤抑制率均大于32 %；⑵与空白对照组相比，生兴克癌冲剂20、10 g/ kg能显著地延长移植小鼠白血病P388小鼠的存活天数(P.01)，生兴克癌冲剂20、10 g/ kg对荷白血病P388小鼠生命延长率均在50%以上；⑶与S180荷瘤组相比，生兴克癌冲剂对荷瘤鼠的免疫器官重量、胸腺指数和脾指数有一定的提高趋势，其中生兴克癌冲剂5 g / kg组对荷瘤小鼠的胸腺指数有一定的提高作用(P.05)；⑷与S180荷瘤组相比，生兴克癌冲剂20、10 g /kg组可提高由荷瘤引起的小鼠血清半数溶血素值的降低(P.01)，表明其可提高荷瘤小鼠体液免疫功能；⑸与对照组相比，生兴克癌冲剂20 g /kg组可提高荷S180肉瘤小鼠的免疫吞噬系数α值(P.01)，表明其可提高荷瘤小鼠细胞免疫功能。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

The results indicated that the supramaximal exercise could not affect the immunoglobulinandinterleukin-1 concentration in blood; in the meantime, immunoglobulin and interleukin-1 concentration showed no significant differences between the long and the short distance players.

本研究获得下列结论：超最大运动不会影响血液中免疫球蛋白与白血球间素-1的浓度；长、短距离选手在超最大运动中的免疫球蛋白与白血球间素-1并没有差异。

It was found that after differentiation the cells expressed the endocrine markers insulin 1, glucagons, somatostatin by RT-PCR, however, these endocrine markers cannot be found in uninduced cells.

通过免疫细胞化学染色可以检测到在ICCs中存在有胰岛素、胰高血糖素、生长抑素和pdx-1阳性的细胞，通过RT-PCR可以在mRNA水平检测到胰岛素、胰高血糖素、生长抑素、pdx-1、ABCG2、葡萄糖转运蛋白等的表达。

The distribution of fibronectin in human oocyte and 2-cell to morulae embryos was also investigated, and the possible roles of fibronectin in fertilization, the development of early embryo and implantation were also discussed.

应用免疫组织化学方法和图像分析技术研究整合素β_3、FN和ER、PR在输卵管粘膜内的表达和变化；采用间接免疫荧光技术对FN在人卵母细胞和早期胚胎内的表达分布进行观察；光镜观察输卵管粘膜上皮细胞的形态学变化。结果：1。整合素β_3在人正常输卵管粘膜上皮纤毛细胞的胞浆内表达。

The present study is involved in the expression of vitronectin , a ligand of integrin, on human spermatozoa and its role in fertilization; the expression of integrin α〓、α〓、β〓 sbunits on human and mouse oocytes and their relationship with maturation of oocytes; the reorganization of cytoskeletal protein, actin, of mouse oocytes mediated with RGD peptide binding with integrins; the roles and mechanisms of integrins in activation of mouse oocytes as trasmembrane signaling receptors, using the methos of cell culture, immunohistochemistry, flow cytometer, laser confocal microscope.

本课题采用细胞培养、免疫组化、流式细胞术、激光共聚焦显微技术进行以下四个方面的研究：整合素配体玻连蛋白在人精子的表达及其与精子功能状态及受精的关系：整合素〓在人、小鼠卵的表达及其与卵的成熟程度的关系：整合素介导小鼠卵内actin骨架蛋白的重组；整合素介导卵激活的早、晚期事件，包括卵细胞内钙离子浓度的变化及孤雌发育胚的形成，并探讨其机制。

LC3-FITC had been used to monitor autophagy by immunofluorescence. It showed that there are a high level of LC3 puncta in HCQ+DOX or BAF +DOX treated cells, and in rapamycin treated cells, moderate level in HCQ, BAF, or DOX singly treated cells, and low level in 3-MA+DOX, and there was diffuse fluorescence in untreated or 3-MA treated cells.

内源性LC3免疫荧光染色显示，羟氯喹+阿霉素、BAF+阿霉素及雷帕霉素处理组MM细胞胞浆可见大量颗粒状荧光，羟氯喹、BAF、阿霉素单用组仅有中等量颗粒状荧光，而3-MA及3-MA+阿霉素组胞浆荧光信号基本呈弥散分布，类似于未处理对照组。5。

This paper dealed with the immunnodulation tests of white wax scale in specific and unspecific immunity. The result showed that the IgM in high dose group was clearly higher than that of the control in serolysin test. Lymphocyte transformation and propagation in mice were notable in high dose group.
