Immunohistochemical localization of inhibin α-subunit in normal mature rats was carried out by using Strept Avidin-Biotin-Peroxidase Complex .The results showed that there was positive immunoreactivity of the McAb to inhibinα-subunit in heart ,kidney , spleen , lymph ,adrenal gland and pancreas cells. It indicated that inhibinα-subunit existed in the above tissues , But no positive sites were seen in hypothalamus, pituitary, lung and liver, that means they have no inhibinα-subunit. Compared with the ovariectomized rats, the hypothalamus and pituitary of the rats injected with inhibin-α fragment(1~32)showed an positive reaction, suggesting that inhibin α-subunit could pass through the blood-brain barrier .
用链霉亲和素-生物素-过氧化物酶(Strept Avidin-Biotin-Peroxidase Complex, SABC)免疫组织化学方法,以抑制素α亚基单克隆抗体对正常雌性性成熟大鼠各组织的抑制素α亚基进行定位观察,结果显示:⑴大鼠心脏、肾、脾、淋巴、肾上腺、胰腺均呈阳性反应,表明存在抑制素α亚基;下丘脑、垂体、肝脏和肺组织呈阴性反应,表明其不存在抑制素α亚基;⑵与去卵巢大鼠相比,外源注射抑制素α(1~32)片段的大鼠其下丘脑和垂体出现阳性反应,说明抑制素α亚基能通过血脑屏障。

Recombinant plasmid pSVH 7 DNA of avian influenza virus H7 subtype heamagglutinin gene was encapsulated with DC-chol/DOPE liposomes and PC/chol/SA liposomes separately. Two-week old SPF chickens were intramuscularly inoculated with 50 μ g/0.2ml of the liposome entrapped PSVH 7 DNA. Four-weeks later, each chicken was challenged with 0.1ml 〓 AIV . One week after the challenge, the secretion of the cloacas was collected and transfected to chicken embryos to isolate the virus. The virus was isolated from 6/6 of the control group, 1/6 of the naked DNA group, 1/6 of the PC/chol/SA entrapped DNA group and 0/6 of the DC-chol/DOPE liposome entrapped group. The HI antibody titers (log2) of the four groups were 6. 83±0.98, 7. 0±1. 26, 7. 83±1. 17 and 8. 00±0.89 respectively 1-week after challenge, and 8. 5±0.55, 8. 17±0.82, 8. 68±0.45 and 9. 33±0.54 respectively 2-week after challenge. The results showed that inoculation of liposome entrapped DNA significantly enhanced resistance to virosis in animals.

Important advance was made to Celastrus and ceangulin V after 20 years studies, but the target of Celangulin V was not verified and the metabolic dynamic of Celangulin V in organism was not touched because of the technique difficulty in testing and analysis of trace Celangulin V in organism. It will solve the difficulty that establishing the immunoassay of Celangulin V. The immunoassay of Celangulin V including the synthesis of artificial antigen, the preparation of antiserum, the immunoanalysis location of Celangulin V and the quantitative immunoassay of Celangulin V were studied systematicly in this paper.

objective effects of plant tannins,chebulinic acid and tellimagrandin i on chemically induced hemoglobin synthesis in k562 cells were investigated.methods the hemoglobin synthesis situation was assayed with benzidine staining.erythroid antigens glycophorin a expression on the surface of k562 cells was labeled by direct immunofluorescence using fluorescein isothiocyanate-conjugated anti-gpa antibodies.then flow cytometric analysis was performed to detect gpa expression levels on surface of the cells.results both chebulinic acid and tellimagrandin i could inhibit the hemoglobin synthesis of butyric acid and hemin-treated k562 cells in a concentration-dependant manner.however,the ba-induced k562 cells were more sensitive to two tannins than heroin-induced cells.conclusion chebulinic acid and tellimagrandin i have inhibitory effect on the erythroid differentiation.
目的 研究可水解单宁诃子酸和特里马素i对氯化高铁血红素和丁酸钠诱导k562细胞红系分化的影响。方法四甲基偶氮噻唑蓝法分析诃子酸和特里马素i对k562细胞生长的影响,联苯胺染色法检测血红蛋白合成情况,应用免疫荧光和流式细胞术检测血型糖蛋白a在细胞表面的表达。结果诃子酸和特里马素i在0.04~0.09?mmol/l浓度下均可显著抑制k562细胞的生长增殖。2种单宁化合物(0.002~0.01?mmol/l)可显著抑制丁酸钠诱导的k562细胞血红蛋白合成,2种单宁化合物(0.01~0.05?mmol/l)还可显著抑制氯化高铁血红素诱导的血红蛋白合成,特里马素i(0.01?mmol/l)还抑制丁酸钠诱导的gpa在细胞表面表达。结论诃子酸和特里马素i对红系分化有抑制作用。

Results:①The amount of human colon carcinoma cell line SW480 treated by quercetin decreased. The morphology of partial SW480 cells was shrunk volume, integrated cell membrane, condensed cytoplasm, pyknotic chromatin, nuclear fragmentation. Apoptotic Corpuscles were found by electron microscope.②MTT colorimetric assay showed quercetin inhibited the growth of human colon carcinoma cell line SW480 in a time- and dose-dependent manner when the concentration of quercetin was 30、60、90μmol/L.③Flow cytometry analysis showed the cell cycle of SW480 cell was restricted in G1/S. G0/G1 phase rate increased and S phase rate decreased with increasing concentration of quercetin and time lasting.④ Zymogram analysis assay showed the secretion of matrix metalloproteinases in human colon carcinoma cell line SW480 treated by quercetin decreased. With increasing concentration of quercetin, the secretion of MMP-2 and MMP-9 decreased.⑤Immunohistochemistry method demonstrated the position expression of Cathepsin-D in SW480 cell was suppressed by quercetin in a time- and dose-dependent manner.
研究结果:经槲皮素处理的人结肠癌SW480细胞数量减少,部分细胞体积缩小,细胞膜完整,胞浆浓缩,核染色质固缩,细胞核碎裂,形成凋亡小体;MTT法检测显示当作用浓度为30μmol/L~90μmol/L时,槲皮素对人结肠癌SW480细胞的生长有抑制作用,其抑制作用随着作用浓度的增加和作用时间的延长而增强;流式细胞学发现槲皮素主要作用于人结肠癌SW480细胞周期的G1/S期,大部分细胞被阻断于S期,随药物浓度的升高和作用时间的延长,G0/G1期细胞比例逐渐增加,S期细胞比例逐渐减少;酶谱分析法检测显示不同浓度的槲皮素能够抑制人结肠癌SW480细胞分泌MMP-2及MMP-9,随浓度的升高,MMP-2及MMP-9的分泌量减少;免疫组织化学法显示不同浓度的槲皮素处理人结肠癌SW480细胞后,Cathepsin-D的表达随药物浓度的升高和作用时间的延长而降低。

METHODS: Female BALB/c mice were treated with CCK-8 and keyhole limpet haemocyanin, splenocytes were acquired and incubated in vitro with KLH restimulation, the productions of Th1 cytokines, interferon-γ, interleukin-2 and Th2 cytokines, IL-4, IL-5 in cell culture supernatants were detected by enzyme-linked immunosorbnent assay. The mRNA expressions of IFN-γ, IL-2, IL-4 and IL-5 in splenocytes were detected by reverse transcription-polymerase chain reaction. The levels of Th1 antibody IgG2a and Th2 antibodies IgG1 in serum were also detected by ELISA.
