Significant Spearman rank correlations were found between the results obtained by Luminex assay and by chemiluminescence immunoassay or time-resolved fluorescence immunoassay. Only 1 microliter of serum and 3 hours were needed for Luminex assay. With the advantages of unique capability of multiplexing and high-throughput, good reproducibility, increased detection range and sensitivity, Luminex assay would have a great potential in clinical application.

本研究应用液相芯片技术利用双抗体夹心免疫分析原理同时检测了人血清中的癌胚抗原、甲胎蛋白和乙肝表面抗原，评价了该方法的线性范围、最低检测限、精密度、灵敏度、特异度、准确度等指标，并对检测结果与化学发光免疫分析法（chemiluminescence immunoassay，CLIA）和时间分辨荧光免疫分析法（time-resolved fluorescence immunoassay，TRFIA）进行了相关性分析。

Part Ⅱ Objective: To study the immunoprotective and the immunotherapic effects of the new nucleic acid vaccine of the hepatitis B: pVAX1-C3d-S2S; respectively in normal and HBV transgenic mice after being inoculated intra- muscularly with this Eukaryotic Expression Plasmid, such as the in situ production of the expressed protein in muscle tissue section and specific humoral immune response in normal mice、the variation of specific antigen、antibody and HBV DNA, especialy the changes of specific antigen in liver.

第二部分新型乙肝核酸疫苗pVAX1 C3d-S〓S免疫预防效应和治疗作用的研究目的：研究含有新型分子佐剂补体C3d的HBV adw血清型的乙肝核酸疫苗重组真核表达质粒pVAX1-C3d-S〓S接种正常BALB／c小鼠和HBV转基因小鼠后，其免疫预防作用（接种局部肌肉中特异性抗原蛋白的表达和所诱生的体液免疫应答规律）以及免疫治疗作用（血清中特异性抗原的表达、特异性抗体的产生、HBV DNA的消长和肝组织中抗原表达的变化）。

Results ①Compared to sham group, ISH and IHC of rat hypothalamic CRHshow positive cells numbers increase, positive areas enlarge and average gray scaledecreases, which reveals the expression of hypothalamic CRH neurons enhances afterPX;②The enhancement of ISH and IHC of hypothalamic CRH in PX rats aftermelatonin supplementation dose-dependently recovers, but it can't return to be normaleither in high dose group or in low dose group and either at 4th week or at 8th week;③IHC of CRH in median eminence of neurohypophysis shows positive areas enlargeand average gray scale decreases, which reveals the releasing of CRH fromhypothalamic CRH neurons increases;④ELISA of CRH in serum shows thedifferences of CRH concentrations among all groups are not significant, which indicates PX and melatonin supplementation have no significant effect on CRH levels.

结果： ①松果体摘除后大鼠下丘脑室旁核CRH 原位杂交和免疫组化的结果与假手术对照组相比均显示阳性细胞数增多、阳性面积增加、平均灰度降低，提示松果体摘除后下丘脑CRH 神经元的表达增强；②松果体摘除大鼠补充褪黑素后，下丘脑CRH 原位杂交和免疫组化染色增强呈剂量依赖性恢复，不过不论是低剂量组还是高剂量组，也不论是术后4 周还是术后8周，均不能恢复至正常；③松果体摘除后神经垂体正中隆起CRH 免疫组化结果显示阳性面积增加、平均灰度降低，提示下丘脑CRH 神经元释放CRH 增加；④ELISA 法检测血清CRH结果示各组血清CRH 浓度差异没有显著性，表明松果体摘除及补充褪黑素对血清CRH 水平没有显著影响。

The result indicated that the percentage of lymphocyte were decreased, the percentage of neutrophile granulocytes were magnified significantly with the lighting time increasing, which was due to the level of the melatonin inhibited in the long time lighting; and to supply different exogenous dose, induce the percentage of lymphocyte were magnifying, the percentage of neutrophile granulocytes were decreased. And the statistical analysis showed there was significant difference.

结果表明：在给予不同光照时间的条件下，随着光照时间的延长，褪黑素的分泌水平受到抑制，导致具有特异免疫功能的淋巴细胞百分率下降，具有非特异免疫功能的中性粒细胞百分率上升，经统计分析表明不同组间的测定结果均存在显着差异；采用24h全天光照，用以模拟机体切除松果腺的情况，再给予一定剂量外源性褪黑素，导致机体内具有特异免疫功能的淋巴细胞百分率上升，具有非特异免疫功能的中性粒细胞百分率下降，经统计分析表明不同组间的测定结果均存在显著差异，并且高剂量褪黑素对机体的影响更为明显。

In this article, we used automotive immunohistochemical stainer to check the expressions of HBsAg and HCV antigens in 100 cases of HCC and their adjacent tissues, mean while, we checked their histological activity index and hepatic fibrosis degree. We also used radioimmunoassay to detect serum levels of 4 serum fibrosis markers and serum AFP levels in these cases. Immunofluorescence labeling and flow cytometry were used to check the T-cell subpopulations and the activity of NK cells of these 100 cases. Then the correlations among these factors were studied.
