尘螨标准化变应原疫苗免疫治疗是一种有效、安全的治疗常年性变应性鼻炎的方法，可能通过减少机体IL-4、IL-5的生成，从而减轻IL-4、IL-5对常年性变应性鼻炎患者的免疫毒害作用，发挥其调节免疫的作用，进而达到有效治疗常年性变应性鼻炎的作用。

Small intestinal biopsies showed subepithelial collagen deposition with varying degrees of villous atrophy and varying numbers of intraepithelial lymphocytes. Four patients had previous biopsies showing enteropathic changes without collagen deposition. Seven cases were associated with collagenous colitis and 1 also had features of lymphocytic colitis. Three patients also had collagen deposition in gastric biopsies. One case was associated with lymphocytic gastritis. Celiac disease (CD, gluten-sensitive enteropathy) was documented in 4 patients. Five patients made a clinical improvement with combinations of a gluten-free diet and immunosuppressive therapy. Two patients died of complications of malnutrition and 1 of another illness. Clonal T-cell populations were identified in 5 of 6 cases tested. Four of these patients improved clinically after treatment but 1 has died. Collagenous sprue evolved on a background of CD in 4 cases. There was no history of CD in others and these cases may be the result of a biologic insult other than gluten sensitivity.

小肠活检表现为上皮下胶原沉积，绒毛萎缩程度不一，上皮内淋巴细胞浸润数量不等。4例患者以往曾做过活检，表现为其他肠病改变，并没有胶原沉积。7例患者合并胶原性结肠炎，且1例还有淋巴细胞性结肠炎的特征。3例患者胃活检也发现有胶原沉积。1例患者合并淋巴细胞性胃炎。4例患者有乳糜泻（CD，谷蛋白敏感性肠病）病史。5例患者经无麸质饮食和免疫抑制治疗后，临床症状有所改善。2例患者因营养不良合并另一种疾病而死亡。6例患者做了T细胞受体基因重排，其中5例发现有克隆性T细胞群，这5例中有4例治疗后临床症状加重，且1例死亡。4例患者在CD的基础上病发胶原性口炎性腹泻；其余患者无CD病史，他们的发病也许是生物源性损害，而非谷蛋白敏感。

We should keep eyes on the human cases of avian influenza virus an its variation for the H5 isolates in Shanghai also has the possibility to infect human directly and the infection of H5, H9 viruses had previously occurred in the population especially in those contacting with animals.

综合上述分析结果，我们认为上海地区人群虽然对原有的H3亚型参考病毒株有较高的免疫保护，但H3亚型病毒HA已出现新的抗原漂移，人群现在对新变异株的保护水平尚未知，有可能在未来引起局限性暴发甚至地方性流行；人群对H1亚型流感病毒免疫保护水平低，同时H1亚型流感病毒株部分与原推荐疫苗株抗原性差别较大，并在现推荐疫苗株基础上继续变异，需警惕发生流行的可能性；H5N1亚型高致病性禽流感地域范围扩大，人间病例不断发生，存在演变成大流行株的威胁，上海市H5亚型禽流感病毒株存在跨宿主屏障感染人类的可能性，而且已发现人群尤其禽、畜类动物接触人群中存在H5、H9亚型流感病毒的既往感染，应警惕禽流感病毒的变异和人间病例的发生。

ZCH-2B8a (IgG2a )是作者研究室最近采用髓细胞性白血病细胞系KG1a作为免疫原自行研制的鼠抗人造血细胞分化抗原的单克隆抗体，已提交第8届国际人类白细胞分化抗原协作组会议（HLDA8）鉴定。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

METHODS:The gene of human ET1 was synthesized according to the preferential codons of E. coli, cloned into the EcoRI and SalI sites of vector pThioHisA. The recombinant plasmid pThioHisA-ET1 was constructed , sequenced and transformed into E.coli TOP10. Induced and expressed fusion protein were identified and analysed by 12% SDS-PAGE and densitometry analyses. After the elution, denature and renature, the fusion protein Thioredoxin-ET1 was obtained by ProBondTM chromatogragraphy. The purity of Thioredoxin-ET1 was detected by HPLC. Inoculate Thioredoxin-ET1 once per mouse every 2 weeks in 25, 50 and 100μg separately on 3 groups for 4 times. 10 days after last inoculation, we obtained venipuncture blood.

根据人ET1的多肽序列合成ET1基因，将其插入到pThioHisA的EcoRI和 SalI位点，重组质粒pThioHisA-ET1进行酶切鉴定及序列测定验证后转化TOP10，IPTG诱导的重组菌经SDS-PAGE检测融合蛋白Thioredoxin-ET1的表达量；表达的融合蛋白用ProBond亲合层析纯化并经HPLC鉴测其纯度；每只小鼠按25、50、100ug/次剂量的Thioredoxin-ET1每两周免疫一次，共4次，最后一次免疫10d后制备抗血清，经Western blot和ELISA检测证明Thioredoxin-ET1融合蛋白具有ET1免疫反应原性。
