Three expression vectors containing mutant gfp which were truncated from 3′ termine were constructed in the present paper.The mutation gfp with different length under the control by T 7 promoter were expressed in E. coli BL21 cells.

本文将突变型 gfp3′末端不同程度地截短，构建成表达载体，在大肠杆菌 BL 2 1中表达了不同长度的绿色荧光蛋白，当其生色基因中 65位 Ser点突变成 Thr，则 GFP的激发波由 396nm增至 4 70 nm，不用 UV激发，仅在自然光源下就可看到强烈的绿色荧光，发光强度增加 6倍，若从 3′端截短 12个氨基酸密码，对发光无影响，但截短 75个氨基酸密码，则激发波长降到 360 nm，发光强度减少 12倍。

This study used the pick-up head from the commercial DVD-ROM as the basis for optical path, and drilled a 2mm hole on the focusing object lens. After combined with the design of a beams plitter, the laser beam source of the two DVD formed two in-line laser beams. To meet the needs, a novel low-noise fiber laser with compound cavity structure is designed. The stable single-mode operation is realized by using a transient self-written fiber grating, produced by the effect of saturable absorption within a section of unpumped erbium-doped fiber, as narrow band filter for mode selecting and linewidth squeezing. A detection method for welding lines based on K-L algorithm is discussed. Infrared light source is applied to get high quality images of welding lines through CCD. Image template libraries for unwelded and welded lines are constructed and the welding line image is processed by K-L algorithm to match with the template libraries to judge if the welding line has been welded. The experiment shows that above method is effective with high precision and speed and it can be applied on real-time detection of welding lines.

提出计算机对所得图像进行滤波和图像分割，讨论了K-L算法的原理及其在红外焊缝图像检测中的应用；以红外光源照射焊缝，通过CCD摄取焊缝的图像，建立已焊和未焊的图像模板库，采用K-L算法对焊缝图像进行处理，并与模板库进行比较，得出焊缝是否焊接的结论；试验结果表明，该方法检测精度高，检测速度快，可以实时运用于对手工电弧焊的焊缝质量检测中。
