本研究主要目的是在建立一个适用於银行消费性贷款之违约风险评估模型，以国内某一家金融机构的某一地区资料为对象，资料内容为已申请消费性贷款之申请人资料。

The objectives of this study are:(1) To amplify Tb wbbL gene encoding rhamnosyl transferase bypolymerase chain reaction from the genomic DNA ofM.tuberculosis H37RV strain;(2) To clone PCR product of Tb wbbLgene into a cloning vector pMD18-T for sequencing;(3) To subcloneTb wbbL gene to an expression vector pET16b to construct pET16b-Tb wbbL; and to overexpress Tb WbbL protein in E.coli BL21(DE3)under different induction conditions;(4) To establish theco-expression system for expressing chaperons of pKJE7 plasmidand soluble Tb WbbL protein in E.coli BL21(DE3) under differentinduction conditions;(5) To test expressed WbbL protein by SDS-PAGEand Western blot methods.

本论文的目的是：(1)利用 PCR 方法从结核分枝杆菌 H37Rv 菌株的基因组DNA 中扩增出 Tb wbbL 基因；(2)将 Tb wbbL 基因克隆到pMD18-T 克隆载体中，经 DNA 序列测定证实为正确的基因；(3)将 Tb wbbL 基因亚克隆到 pET16b 表达载体中并通过改变不同的诱导条件在大肠杆菌 BL21(DE3)中表达 Tb WbbL 蛋白质；(4)建立在 BL21(DE3)大肠杆菌中共表达 Tb WbbL 蛋白质与分子伴侣的体系，优化表达条件以高效表达出可溶性 Tb WbbL蛋白质；(5)用 Western blot 方法鉴定所表达的 Tb WbbL 蛋白质为 Tb wbbL 基因产物。

The purpose of this study is to explore the trend of CO2 emission in the electricity industry. An input-output analysis and FGP along with the IPCC method are employed to estimate the volume of CO2 emission produced from power sources in the electricity industry from 1996 to 2006, focusing on the correlation between derived energy and carbon dioxide. The potential volume of CO2 emission in Taiwan between the years 2007 and 2015 is also estimated according to the information on the government's long-term development program. Furthermore, the input-output approach and FGP model is constructed to draft future CO2 reduction program for the electric power industry. The program includes fuel substitute of dynamotors and the promotion of electrical efficiency; at the same time, analysis is made on the correlation between CO2 and GDP in this reduction program.

摘要 研究目的为探讨电力事业中二氧化碳排放趋势，以投入产出分析法及模糊目标规划法搭配IPCC方法，推估1996~2006年间整体产业各能源二氧化碳排放量，重点为次级能源电力与二氧化碳间的关联性，并依据我国长期开发方案所提供资料推估我国2007~2015年间二氧化碳排放量，并进一步建构投入产出模糊规划模型，拟定未来电力事业可行的二氧化碳减量方案，方案包括发电机组燃料替代与电力效率提高两种，并且分析减量方案中产业二氧化碳与GDP两者间的关系。

Objective:The aim of the experiment is to search for the regulation that the hepatic stem cells abnormally differentiate into cancer cells. In the experiment,the variating regulation of PCNA,c-kit and c-myc in the formation and development of HCC will be interpreted with immunohistochemistry and RT-PCR. So,the initial mechanism that hepatic stem cells abnormally differentiate into cancer cells from the levels of cell,protein and gene will be clarified. The last,the inhibition to HCC with ulinastain will be studied,from which,a new way of clinical prevention and treatment for HCC will expect to be explored.

二、目的本实验旨在探讨以卵圆细胞为代表的肝干细胞在HCC发生、发展过程中在肝组织内分布规律，并通过免疫组化、RT-PCR等技术阐明PCNA、c-kit、c-myc在HCC发生、发展过程中的动态演变规律，从细胞、蛋白、基因三个水平初步研究卵圆细胞向癌细胞异常分化的机制，同时观察乌司他丁对HCC发生、发展的抑制作用，为肝癌的临床预防和治疗探索新的途径。
