Studies on sequence changes in the early genomes of interspecific F_1 hybrid and amphidiploid in CucumisIn the present study,extensive genomic changes were detected in newly obtained allotetraploid of Cucumis,namely Cucumis hytivus Chen and Kirkbride,by AFLP analysis using 12 pairs of EcoR-I/Mse-I primers.

甜瓜属远缘杂种F_1及人工异源双二倍体早期基因组序列变化研究利用12对Eco RI/Mse I选择性引物，对甜瓜属人工异源双二倍体C.hytivus及其二倍体父母本进行AFLP分析，结果发现多倍体基因组在形成早期发生了广泛的序列变化。

The structure and expression pattern of each storage protein was investigated using one-dimensional SDS-PAGE. Our results showed that the protein content of each fraction in endosperm of autotetraploid rice line is mostly higher than that of its corresponding diploid rice line.

结果表明：同源四倍体水稻胚乳蛋白各组分含量与相应的二倍体相比，大部分呈增加趋势；同源四倍体水稻胚乳蛋白的亚基类型与相应的二倍体水稻基本一致，仅在全蛋白电泳和清、球蛋白电泳中各发现1条差异条带。

Tetraploid embryos could be produced by electrofusion at the stage of two-cell embryos, which could develop to blastocysts followed by fusion of cytoplasm and nucleus and cleavage in vitro. During the fusion of cytoplasm, the DNA methylation levels of the fused embryos are as high as these of two-cell diploid embryos in vivo Then the embryos are rapidly demethylated when the nucleus begin to fuse, resulting in the lowest DNA methylation levels when the nucleus are fused completely. After that, the DNA methylation levels of the fused embryos are gradually increased until the morula stage. However, whereas an asymmetric distribution of DNA methylation is established in vivo-derived blastocysts with a higher methylation level in the inner cell mass than that in the trophectoderm, we can not detect the asymmetric distribution in most in vitro-derived tetraploid blastocysts.

结果表明：利用电融合方法制备的小鼠四倍体胚胎在体外培养体系中经历细胞质融合、细胞核融合及细胞继续分裂发育直到囊胚期的过程，在细胞质融合的时候胚胎卵裂球同体内体外培养二倍体胚胎一样，呈现高度甲基化状态；在细胞核开始融合的时候，甲基化水平急速下降，在细胞核完全融合的时候甲基化水平达到最低点；随着胚胎继续分裂，胚胎甲基化水平逐渐增加，在桑葚胚期甲基化水平最高；但是囊胚期四倍体胚胎内细胞团同滋养层细胞甲基化荧光信号没有差别，这与体内体外培养二倍体囊胚内细胞团细胞甲基化荧光强度高于滋养层细胞甲基化荧光强度不同。

Using semi-quantitative RT-PCR, the differential expression profiles of eleven selected genes were confirmed in the ovaries of triploid and diploid. These genes fell in gene categories with a wide range of functions. The results indicated that triploidy affects the dynamic gene regulatory network in triploid ovary. This study established a firm basis for future investigation on characterization of crucial molecular events for normal ovarian development in shrimp.To further dissect exact gene functions for gonad development of shrimp, three differentially expressed genes between diploid and triploid ovary, PCNA (proliferating cell nuclear antigen), CAS/CSE1 (cellular apoptosis susceptibility protein/chromosome segregation 1) and SSRF (spermatogonial stem-cell renewal factor) were characterized on certain aspects.

The genetic analysis and improvement of characters, such as tuber processing quality traits and yield components which tend to be quantitatively inherited, are far more difficult than those at diploid potato. Reviewing the earlier studies indicated that a few systematic investigations on genetics of important traits in potato were untaken and most focused on estimates of combing ability for agronomic traits due to limited materials and methodologies available.

In order to explain clearly the variation status of embryo from tetrapoid Robinia pseudoacacia and fully use the variation resource initiated in the sexual reproduction of TRP in the further improvement of TRP, the classification and vitalities of seed embryos from tetraploid Robinia pseudoacacia were studied. The main results are as follow. According to the cotyledon number, color and plumpness, the seed embryos of TRP could be divided into 5 types: YVT (yellow, very replete,2 cotyledons), YMT( yellow, more replete,3 cotyledons ), YRF ( yellow, replete,4 cotyledons ), GNT ( green, no-replete, 2 cotyledons ) and WNT ( white, no-replete, 2 cotyledons ).

为了阐明刺槐同源四倍体种子胚变异状况及充分利用刺槐同源四倍体有性过程创造的变异资源对四倍体刺槐进行遗传改良，研究了刺槐同源四倍体种子胚类型及其生活力特征，主要结论如下：刺槐同源四倍体种子胚按照子叶数量、颜色及饱满程度，可以划分出5个类型：黄二、黄三、黄四、绿二及白二，所有类型种子胚混合平均单胚重量接近二倍体的1/2。

The result showed that: 1 Gill epithelium nucleus major-axis size, hemolymph nucleus diameter in the connective tissue of mantle and labial palps were significantly (P＜0.001) larger in triploids than in diploids, no significantly differences were found in rectum epithelium height between diploids and triploids.
