乙胺

EMB-resistance in 18 isolates was caused by the mutation at coden 306 in embB gene with positive rate of 62.1%. The PCR result was negative in 20 specimen from pulmonary diseases without active TB and 5 negative control strains of no MTB. The data in this study showed high specificity of 100%. Only one specimen in 7 specimen with negative result in acid-fast stain and cultivation was shown alteration in coden 306 in embB gene.

结果：130例活动性肺结核患者痰标本抗酸染色阳性38例，阳性率为29.2%(38/130)；罗氏培养阳性为79例，阳性率为60.8%(79/130)。79例培养阳性痰标本中耐乙胺丁醇29例，其中有18例在embB基因met306ATG突变，基因突变率为62.1%。20例非结核肺部疾病患者痰标本和5株非结核分枝杆菌菌株经套式PCR扩增全部为阴性，特异性为100%。7例涂阴培阴PCR扩增阳性的序列中有1例在embB基因306位密码了发生突变。

Methods The internal standard was diazepam, the samples were extracted by ethyl acetate after basified the plasma., the column was Hypersil C18, and the mobile phase mixed of methanol: water(75:25) containing 0.03%-triethylamine.

色谱柱为Hypersil ODS C18 4.0mm×250mm，流动相为含0.03%三乙胺的甲醇：水（75：25），流速1.0ml/min，紫外276nm波长检测。

In the orthogonal designable experiment ,the analysis results of zirconium source, precipitation , the sulfating agent is 1.54%, 2.27%, 5.11%,respectively. This result show that the influence of sulfating agent to the catalytic activity is the biggest. Under the same density of the sulfuric acid soaks, the effect of sulfuric acid is well compared to ammonium sulfate .The second is the precipitation, but the influence of the zirconia source is the smallest.

正交设计实验中锆源、沉淀剂、硫酸化试剂的极差分析结果分别为1.54%，2.27%，5.11%，此结果说明硫酸化试剂对催化剂的影响最大，其次是沉淀剂，锆源的影响最小，同时表明以Zr(NO3)4·5H2O为锆源、乙胺为沉淀剂、硫酸为硫酸化试剂是催化剂的最佳制备方案。

The optimal conditions determineded by experiments were as follow: chloracetic acid : sodium carbonate : isopropyl xanthate : ethylamine=l : 0.58 : 1 : 1.05, water usage is 30 ml/O.lmol chloracetic acid, esterification temperature was 80癈, esterification time was 2h, ammonolysis temperature was 70 C, ammonolysis time was Ih.

实验得出最佳工艺条件为：一氯乙酸：碳酸钠：异丙基黄药：乙胺=1：0.58：1：1.05，水用量为30 ml／0.1mol一氯乙酸，酯化温度80℃，酯化时间2小时，氨解温度70℃，氨解时间1小时。
