Evaluation of IEM103 in rabbit animal model.The evaluation was based on the control-challenge study (IEM101 as control). Rabbits were laparotomized, injecteded 10〓 cfu directly into proximal small bowel, and 28 days later, challenged with 0.25--2ug CT and 10〓-10〓 cfu 4 wild toxic strains with different biotypes and serotypes. Experimental group developed antitoxic and significant vibriocidal immune responses to vaccination, and could protect no less than 2ug CT and 10〓-10〓 cfu all wild strains challenge.

IEM103的免疫原性评价：经兔肠段结扎免疫〓cfu IEM103，7天后在外周血中可检测到外周淋巴细胞分泌性抗CT IgG，血清中可检测到抗CT抗体、杀弧菌抗体，P／N均≥2，且可持续至28天。28天后用0.25-2ug CT毒素，四种毒株滨-43，吴江-2，395，1119〓cfu菌量攻毒，实验组可保护至少2ug CT攻毒，而对照组可保护1ug以下CT攻毒，两组均可保护〓cfu四种毒株攻毒（以肠积液量≥1.0ml／cm为不保护判断标准）。

Results: The bleeding site: in 136 cases, 66 cases of upper gastrointestinal bleeding (48.5%), 42 cases of lower gastrointestinal bleeding (31%), all 28 cases of gastrointestinal bleeding (20.5%); bleeding: 34 cases of bleeding ( 25%), amount of bleeding in 70 cases (51%),32 cases of heavy bleeding (24%); the cause of bleeding: 87 cases of stress ulcer (64%), neonatal necrotizing enterocolitis 25 cases (18%), 16 cases of neonatal hemorrhagic disease (12%), intestinal malrotation volvulus merge two cases (1.5%) and stomach wall muscle dysplasia and 2 cases (1.5%), 4 cases of unknown causes (3%); Treatment Results: 129 cases of cure (95%), the death of 4 (3%), three cases of auto-discharge (2%).

结果：出血部位：136例中，上消化道出血66例（48.5％），下消化道出血42例（31％），全消化道出血28例（20.5％）；出血量：少量出血34例（25％），中量出血70例（51％），大量出血32例（24％）；出血的病因：应激性溃疡87例（64％）、新生儿坏死性小肠结肠炎25例（18％）、新生儿出血症16例（12％）、肠旋转不良合并肠扭转2例（1.5％）和胃壁肌层发育不良2例（1.5％），原因不明4例（3％）；治疗结果：治愈129例（95％），死亡4例（3％），自动出院3例（2％）。

Methods From Feb 2006 to Jun 2006,188 hospitalized children in Shenzhen children s hospital, were collected deep tracheal aspirate at the time of hospitalization. The respiratory tract secretions were immediately sent for bacterial culture with 3 kinds of medium:ordinary medium, Hemophilus influenzae selective medium, Streptococcus penumoniae selective medium. Then we extracted the total nucleic acids from secretions, and detected Mycoplasma pneumoniae by single fluorescent quantitation PCR. Simultaneously, 14 respiratory tract pathogenic bacterium and Mycoplasma pneumoniae were detected by Target Enriched multiplex PCR. Amplification products were identified by the Luminex100 suspension array.

确诊为社区获得性肺炎的患儿188例，在入院当天采集深部呼吸道吸引物，用普通培养基和肺炎链球菌、流感嗜血杆菌选择性培养基进行细菌培养，然后提取深部呼吸道吸引物中病原体的DNA，采用荧光定量单PCR的方法检测肺炎支原体，并对同一标本采用靶序列富集多重PCR技术同时扩增肺炎链球菌、流感嗜血杆菌、金黄色葡萄球菌、肺炎克雷伯菌、大肠杆菌、嗜肺军团菌、铜绿假单胞菌、鲍曼氏不动杆菌、脑膜炎奈瑟氏菌、阴沟肠杆菌、奇异变形杆菌、化脓链球菌、粪肠球菌及屎肠球菌14种呼吸道病原菌和肺炎支原体的靶基因，扩增产物用Luminex100多功能悬浮点阵仪检测。

And more, ERK contents between group A and B, group C and D were not dramatically different. The pathological lesions indicated that intestinal tissue necrosis was severe in group C and D, which were graded 3 points, but obviously lessened in group A and B, which was graded 1 point, with ITF interfering. Conclusion: Intestinal inflammation was ameliorated after ITF was injected hypodermically or intraperitoneally.

结果：A、B组组织匀浆中ERK的面密度值各为87.68±19.24、82.65±21.35，较C、D组(30.23±7.79、30.74±8.19)明显升高(P＜0.01)，C、D组与E组(5.41±1.46)比较也有升高(P＜0.05)；并且随著ERK的激活伴随著明显的核转位。A、B组间及C、D组间ERK含量无显著差异(P＞0.05)；病理切片显示C、D组HE染色切片见肠壁损伤轻重不一，可见全肠粘膜绒毛坏死，病理评分的中位积分为3分；A、B组肠上皮细胞少量脱落，顶端绒毛坏死，病理评分的中位积分为1分。

Activation of NF-κB(p65) with evident nuclear translocation was found in neonatal rat NEC model. The pathological lesions indicated that intestinal tissue necrosis in varying degrees in group C and D, which were 2-4 points, but obviously lessened in group E, which were 0-2 points. Conclusions Intestinal inflammation is ameliorated after the ITF is injected hypodermically.

结果 C、D组组织中NF-κB(p65)阳性表达较A、B及E组显著升高P(下标 a.01，E组与A、B组比较也有升高P(下标 a.01，且随NF-κB(p65)的激活有明显的核转移；病理切片示C、D组HE染色见肠壁损伤轻重不一，可见全肠黏膜绒毛坏死，病理评分2~4分；E组轻度肠上皮细胞脱落，顶端绒毛坏死，肠组织病理评分0~2分。
