Methods: The analytical column was SHIMADZU VP-ODS C18 column(4.6 mm×150 mm 5 μm) with a mobile phase consisted of 0.05 mol/L disodium hydrogen phosphate solution(85% phosphoric acid solution with pH adjustment to 2.5)-acetonitrile-methanol(42.5∶47.5∶10) at the flow rate of 1.0 ml/min the detection wavelength was set at 254 nm.

目的：建立丙酸氯倍他索尿素乳膏中丙酸氯倍他索含量的HPLC测定方法。方法：色谱柱为SHIMADZU VP-ODS C18柱(4.6 mm×150 mm5 μm)流动相为0.05 mol/L磷酸氢二钠溶液(用85%磷酸溶液调节pH值至2.5)-乙腈-甲醇(42.5∶47.5∶10)流速为1.0 ml/min 检测波长为254 nm。

本文以竖向支承构件有一定侧向刚度的预应力混凝土框架结构为研究对象，分析了框架梁受到柱的侧向约束时的受力机理，并运用有限元数值模拟计算方法全面分析了随着梁柱线刚度比、跨度、跨数等诸多因素的变化，框架梁中预应力有效量的分布和变化情况，得到了侧向约束对框架梁建立预压应力的影响因素和具体结论。

The principal of buffer is described as follows.At impulsing down,the piston moves down and vacates the annular space.The pressure in respiratory cavity decreases to a certain level and the traveling valve opens.At that time,the liquids enter into traveling valve and cavity and the plunger is impulsed upward to carry out the gentle transient at the sudden change.When the upward impulsing begins,the respiratory cavity is subjected to pressure and the traveling valve closes.During the gradual pressure rising,the liquids in respiratory cavity effuse from the upper ventage and the sudden change of force-summing direction of plunger is relieved relying on the resistance of ventage.

其原理大致是：下冲程时，活塞向下运动，让出环形空间的体积，呼吸腔内压力下降，下降到一定水平后，游动阀打开，此时，游动阀和小孔同时进液，使柱塞在从上死点处突然改变方向向上运动时得到缓冲，实现了柔和过渡；当上冲程开始时，呼吸腔受压，游动阀关闭，在压力逐渐上升的过程中，呼吸腔内的液体从上部的小孔流出，并依靠小孔的阻力使柱塞受力突然换向得到缓冲。

The results showed as follows: 1 Extraction of gelsemium total alkaloids. Total alkaloids were extracted with 95 per cent ethanol. The extract rate was 0.519 per cent and the extract purity were 88.02 per cent and 87.38 per cent; Total alkaloids were extracted with chloroform .The extract rate was 0.416 per cent and the extract purity were 89.02 per cent and 94.52 per cent. 2 Isolation of gelsemium total alkaloids with TLC. Total alkaloids contained five or more alkaloids. The optimal developing agent was chloroform :methanol :ethyl acetate(9:l:0.3),the optimal eluent was methylene chloride:methanol(92:8). 3 Isolation of total alkaloids with column chromatography. Alkaloid A and alkaloid B were obtained, alkaloid B was identified as koumine.

研究结果如下：1、钩吻总碱的提取：用95％的乙醇提取钩吻生物碱提取率为0.519％，其提取物纯度为88.02％、87.35％；用氯仿提取钩吻生物碱提取率为0.416％，其提取物纯度为89.02％、94.52％。2、钩吻总碱的薄层层析分离：总碱中含有5种或5种已上的生物碱；氯仿：甲醇：乙酸乙酯(9：1：0.3)是最佳薄层层析的展开剂；二氯甲烷：甲醇(92：8)是柱层析的最佳洗脱剂。3、钩吻总碱的柱层析分离：共得到两种钩吻生物碱单体成分碱A和碱B，碱B为钩吻素子，碱A可能是钩吻素甲。4、钩吻总碱的急性毒性试验：钩吻总碱小鼠腹腔注射的半数致死量LD_(50是：7.38mg／kg，95％的可信限范围为：6.67mg／kg～7.94mg／kg。
