The differentiation coefficient is 0.07 between three inbreed families respectively. The highest level of genetic homozygosity is in families Ⅱ at 18, but genetic variation is a little difference between inbreeding pig and mice. However, this study revealed the pattern of genetic variation at microsatellite loci in the 3 inbred families of WZSP with one of the most important findings to be the high level of unexpected heterozygosity at some loci, e.g. Sw874 and Sw936 that could be part of the unique genetics of three families. A hypothesis is that certain functional genes linked with such microsatellite loci may be involved in the maintenance of viability of these inbred families of WZSP remains for further investigation.

随近交代数的推进，每个星座上等位基因数越来越少，基因的纯合度越来越高，但WZSP近交系微卫星等位基因的纯合度有一定的限度，与近交鼠有所不同；在近交系3个家系各个世代中有少数几个基因座，如Sw874和Sw936等一直处于高度杂合状态，这可能与WZSP近交家系的种质特异性有关，推测与这些基因座处于同一连锁群的某些功能基因在维持极高近交水平下WZSP的基本生存能力中起着关键作用。3个近交群体间的分化系数在0.07以上，已各自成为独立家系；3个家系的近交程度不完全一致，基因的纯合度亦不同，Ⅱ系最高、14个微卫星座中7个纯合，其次为Ⅰ系和Ⅲ系。

ERAF17 gene expresses differently between male and female reproductive organs. Its expression is induced by ethylene or ethylene released compounds that can promote femaleness. No evidence directly shows the relationship between ERAF17 gene and femaleness, however. So this work focus on proving the positive correlation between the expression of ERAF17 gene induced by ethylene and development of female flowers,then search for the signal pathway of cucumber female reproductive organ development.

ERAF17基因在黄瓜的雌雄生殖器官的表达不同，可以由乙烯诱导表达，从而推动雌性化，但是它的作用机制却不清楚，并没有明确的证据表明ERAF17基因与雌花的发育有直接关系，所以本论文研究重点在于证明乙烯诱导ERAF17基因的表达对黄瓜雌花的分化有确定的正相关，从而可以探索黄瓜雌性器官发育存在的可能的信号通路，并对应用前景进行评估。

Based on the crossing data among these flatfishes, the relationship between pairwise genetic distance and hybrid fitness was evaluated. The findings indicated that positive relationship might exist between genetic distance and hybrid fitness. The pairwise genetic divergence between species decreased as the interspecific hybrid fitness largened. These results implied that speciation clock might exsit in Pleuronectiformes.

Total RNAs from KAx-3 cells and AK127 cells(developed for 14h) were isolated. After the reverse transcription and PCR reaction, two distinct differential fragments were acquired., fragment A was from KAx-3 cells and fragment C was from AK127 cells. After retriving and reamplifying the differentially expressed fragments, white-blue plaqueselection, the fragments were purified. Northern blot proved that fragment A was from KAx-3 cells and fragment C was from AK127 cells. The results of sequencing and researching for NCBI database have been showed: part sequence of fragment A shows 91% similarity to the gene encoding DhkA, 92% similarity to the gene encoding DhkF, 91% similarity to the gene encoding STATc, 97% similarity to the homoeobox gene encoding protein. These genes play important part in controlling cell differentiation and cell proportion in Dictyostelium discoideum.

本研究通过提取盘基网柄菌发育14小时的野生型KAx-3细胞和突变型AK127细胞的总RNA，运用mRNA差异显示法分离出了两条明显的差异表达片段，其中片段A来自野生型KAX-3细胞，片段C来自突变型Ak127细胞；并通过凝胶回收差异片段、对差异片段进行再次PCR、蓝白斑筛选克隆、提取质粒、酶切电泳纯化差异片段；接着进行Northern杂交的结果表明，片段A只与野生型KAx-3细胞的总RNA有杂交信号，片段C只与突变型AK127细胞的总RNA有杂交信号，这就排除了差异片段假阳性的可能；最后通过测序，搜索NCBI BLAST数据库发现：片段A的小部分序列与编码组氨酸激酶DhkA基因中一段序列的相似性高达91%，与编码组氨酸激酶DhkF基因中的一段序列相似性高达92%，与编码STATc蛋白基因的一段序列相似性达91%，以及与编码同源框蛋白的基因中的一段序列相似性达97%，这些基因在盘基网柄菌细胞分化和细胞比例调控过程中起着相当重要的作用，这些数据进一步说明了突变细胞不能完成发育的原因。
