In order to diagnose disease caused by muscovy duck parvovirus rapidly and accurately, and at same time, further study the taxonomic relationship between muscovy duck parivovirus and goose parvovirus, two pairs of PCR primers (LHMP1/LHMP2 and LHGP1/LHGP2) were designed respectively base on complete nucleotide sequences of Muscovy duck and Goose parvoviruses registered in the GenBank, The LHMP1 for the upper one and the LHMP2 for the lower one, corresponding to the specific nucleotides sequence 454-472 and 1173-1155 respectively in MDPV, and the LHGP1 for the the upper one and the LHGP2 for the lower one, corresponding to the specific nucleotides sequence 454-472 and 1173-1155 respectively in GPV.

Next the discussion in this study, about an ellipse with an angle of attack and two circular cylinders with different diameters in the flow, the aim of this study is investigated for the ability of the present numerical method by solving the suppression of the vortex shedding flow behind the two-cylinder system which was kind of difficult to reach by a Cartesian grid method. In this study, the suppression of the vortex shedding flow is tested to solve a uniform flow past through two different diameter cylindrical in the flow field with Fractional Step Method and Immersed Boundary Method.The numerical method which we investigated is showed that the results of the problem is appropriable in other literatures.

接著探讨流过具攻角之椭圆柱体流场及流过圆柱抑制涡漩逸出之流场，目的是要研究利用控制圆柱来抑制下游涡流的流场行为，本文采用分步法配合内嵌边界法数值解析一均匀流流经两不同直径的圆柱流场，来测试抑制涡流逸出的问题，根据现有的数值结果发现试验运用了合适的方法且得到令人满意的结果。

Three major clusters of numt sequences ( cyt b numt、 ATP8+ATP6 numt and ND4+ tRNA-His+ tRNA-Ser numt) were particularity amplified and sequenced from 10 species within Galliformes and A.galericalata. Comparison numts sequences with homologous mitochondrial partial genes get the different evolutionary characteristics and evolutionary speed compared to mtDNA, then examine the age of insertion of numts using the sequence divergence of numt from the mtDNA. Finally, joined Coturnix japonica、 Coturnix chinensis、 Lophura leucomelana、 Lophura swinhoii and A.albifrons mitochondrial sequences in GenBank, we study the importance function to phylogenetic research of mitochondrial genes and homologous numts.The evolutionary rate of Numts is lower than their authentic sequences of mtDNA, cyt b and ND4 gene evolve faster than homologous numts 2.5 times; ATP8+ATP6 gene evolve faster than numts 18.9 times in Galliformes. Numts show less codon position bias and transition bias under not phylogenetic pressure.

根据2004年3月GenBank登录的红原鸡核基因组草图与其线粒体基因组比对发现红原鸡核基因组中存在3段较长的线粒体假基因，利用PCR方法扩增鸡形目10种鸟类和雁形目中鸳鸯核基因组中3段numt序列：cyt b numt、ATP8及下游ATP6 numt和ND4+tRNA-His+tRNA-Ser numt，测序后分别比较线粒体基因与核同源numt序列的进化特点和进化速率，确定numt序列插入核基因组的时间，以及结合GenBank已登录的日本鹌鹑、蓝胸鹑、黑鹇、蓝鹇和1个雁形目物种——白鹅雁(A.albifrons)相应序列探讨两种同源序列对于鸟类系统进化发育重建的重要作用。

PART 2:The molecular mechanisms of Nontypeable Haemophilus influenzae induced inflammatory response in respiratory epithelial cells Objective:Nontypeable Haemophilus influenzaewas traditionally considered as an extracellular pathogen in respiratory infections.In this study,we established a model of infected cells to confirm NTHI was also a facultatively intracellular pathogen.Mitogen activated protein kinasesare the important signal transduction pathway in mediating inflammatory response induced by NTHI in respiratory epithelium.In this study,we elucidate the impact of NTHI,and its SCF and EP on the expression of MAPK and subsequent proinflammatory mediators.

第二部分：NTHI致呼吸道上皮细胞炎症反应的分子机制研究目的：通过建立呼吸道上皮细胞株的体外NTHI感染模型，确认NTHI在细胞内的存活，观察NTHI及其组分(可溶性胞质成分SCF、外膜蛋白EP)对呼吸道上皮细胞MAPK信号传导通路和后续的前炎症因子表达的影响，并应用特异性ERK-MAPK抑制剂U0126、p38 MAPK抑制剂SB203580特异性阻断下游MAPK信号传导通路，观察它们对呼吸道上皮细胞分泌前炎症因子表达的影响。

The N gene ORF was then subcloned into pET-30a vector and the recombinant plasmid was transformed into E.coli BL21 (DE3) and induced with IPTG. The protein expression was determined by SDS-PAGE. The expressed protein had a molecular weight of 54.4 Ku that existed as inclusion body. Thin-layer scanning showed that the expression product accounted for 30.5% of the total bacterial proteins. The recombinant protein possessed native biological activity and could react with anti-PEDV hyperimmune serum in Western blot.

以阳性质粒为模板，用分别含有BamH Ⅰ和Xho Ⅰ酶切位点的上、下游引物扩增得到ORF，其PCR产物经BamH Ⅰ和Xho Ⅰ双酶切后定向克隆到pET-30a载体，构建的重组质粒命名为pET-30a-PN；将pET-30a-PN转化到大肠杆菌BL21 (DF3)中，在IPTG诱导下进行表达；SDS-PAGE结果表明表达出与预期大小相符的约54.4 Ku的重组蛋白，重组蛋白以包涵体形式存在；薄层扫描结果表明表达产物占菌体总蛋白的30.5%；Western blot分析表明表达的重组蛋白能与抗PEDV高免血清反应，说明该重组蛋白具有免疫学活性。
