Methods Both in vitro and in vivo models of ventilator-induced lung injury were used. Alveolar epithelial cells were stretched in vitro to mimic the lung injury in VILI. Rats were ventilated at large tidal volume to produce ventilator-induced lung injury in vivo. A total of 23 inflammatory cytokines were screened with micro gene array in stretched alveolar epithelial cells.

方法通过呼吸机诱导肺损伤的体内及体外模型，将分离的肺泡上皮细胞于体外拉长，利用基因微阵列技术分析并筛选出上调的炎性细胞因子，之后给予大鼠呼吸机机械通气，留取血清标本，测量这些细胞因子在血清中是否上调。

Cytokines found to have up-regulated in cells were measured in serum and lung tissue of rats exposed to large tidal volume ventilation. For investigating the intracellular pathway of cytokine up-regulation in VILI, exogenous TNF-α or H2O2 was added to culture media of alveolar epithelial cells. Cytokines were then measured. To explore the role of oxidant stress in VILI, N-acetylcysteine, as an anti-oxidant, was used in vitro and in vivo.

Compared with the control, the lungs of 3-day-hyperxia-exposure group developed into edema, hemorrhage and inflammatory infiltration and this condition aggravated at the 7th day. The pulmonary septum was significantly widened at the 14th day, the interstitial fibrocytes as well as alveolar epithelial cells proliferated obviously, and the tendency of lung fibrosis was also presented. The results mentioned above showed that the modle of hyperoxia lung injury was successfully established, and the acute inflamationm was its early symptom while the fibrosis was its late manifistation. There was no positive cytes of IFN-γ in the lung of nomal rats, The positive cytes of the groups under hyperxia-exposure for 3, 7, 14 day, which climbed to the climax at the 7th day and began to decline from the 14th day, were found in alveolus epithlia,pulmonary interstitial, bronchoepithlia and macrophage.

高氧14天（5.46±0.30）虽有所下降，但同空气组比，差异仍然具有显著性（P.05）；BALF中蛋白含量高氧3天（0.45±0.10）、7天（0.66±0.08）、14天（0.79±0.14）显著高于空气组（0.18±0.08，P值均。05）；肺通透系数高氧3天（6.61±2.05）、7天（10.88±1.54）、14天（12.75±2.20）均明显高于空气组（2.99±1.27，P值均。05）；同时病理组织学改变也表明与空气组比较，高氧3天时肺组织水肿、出血、炎性细胞浸润，7天时进一步加重，14天时肺间隔明显增宽，间质纤维细胞和肺泡上皮细胞明显增生，出现肺纤维化倾向；以上结果均表明我们成功地建立了高氧肺损伤动物模型，且急性炎症反应是其早期表现，纤维化改变是其晚期表现。

RESULTS:In BN rat retina,Cx43 was immunolocalized to internal limiting membrane,nerve fiber layer,ganglion cell layer and pigment epithelium.Cx43 appeared in the form of distinct puncta distributed on ganglion cell.
