Chicken IL-15(ChIL-15), Which was discovered in 2001, plays a main role in activating NK cells and CD8+ memory T cells, and may therefore have potential as a vaccine adjuvant.According to published ChIL-15 gene sequence, a pair of primers were designed and synthesized to clone ChIL-15 cDNA from ConA-activated chicken splenocytes by RT-PCR. Recombinant plasmid pUC19ChIL-15 carrying the ChIL-15 gene was constructed. The gene encoding mature ChIL-15 (mChIL-15) was amplified from pUC19ChIL-15 by PCR and cloned into pMD 18-T vector. After digested with Kpn I /Pst I , the mChIL-15 gene was subcloned into prokaryotic expression vector pPRoEX橦Ta and transformed into E.coli DH5a .The transformant was induced by IPTG, and the recombinant ChIL-15(rChIL-15) was expressed as a fusion protein with a histidine hexamer tag at the N-terminal end of the protein. Following expression, the protein was purified by ProBond?Nickel-Chelating Resin. The uninduced and induced protein lysates and the purified protein were separated by SDS-PAGE and transferred onto nitrocellulose membrane to identify rChIL-15 by Western blot. The rChIL-15 was used to immune the guinea pig to obtain rChIL-15 polyclonal antibody.

The monoclonal antibody was produced by the method of inoculating BALB/c mice intraperitoneally with the hybridoma cells, and purified with protein A-Sepharose 4B affinity chromatography.3 Establishing of ELISA protocol to detect hBLySTwo kind of different ELISA protocols were tried to study the experiment condition of detecting hBLyS level. Streptavidin was used to indirect coating in protocal 1, but biotin-avidin system was used to enlarge the final reaction signal in protocal 2. The optimal reaction condition was chosen by the experiments. The sensitivity of protocal 1 was poor, but the linear relation of protocal 2 standard curve was excellent when the concentration of hBLyS was between 3.2~400 ng/mL. So a protocal was established which sensitivity can reach to nanogram.

The content of chlorophyll of microwave samples was hig her than steaming samples,and there was a small difference of caffeine content b etween microwave and steam samples.There was a little higher content of theafla vins in microwave samples than steam samples.The content of thearubigins in steam samples was higher than microwave samples,and there was no differe nt theabrown between microwave and steam samples.

This paper reviewed the developments in 2-D numerical model of the water flow. By means of the latest observation data of the prototype, the detailed analysis was made on the major thalweg, section, island and bottomland, the erosion and deposition of the river bed and the scour hole near the dock and the law of the fluvial process was obtained. Based on the related references, the evolution trend of the channel after the Three Gorges was finished was predicted. The 2-D mathematical model of the water flow was set up and validated using the latest data. With the model, the flow and velocity of several cases were calculated. The stability computation of the bank at the typical section under difference conditions was carried out by adopting the world widely used Geo-Slope slope stability analysis program. Finally, the synthetical evaluation on the influence of the project on the flood level, velocity, river circumstances, bank protection and flood prevention was made and the conclusion that the construction of the dock had little effect on the flood discharge was obtained.
