Reducing the complexity of vector quantization is still a main problem. This paper presents a new algorithm of minimum distortion predicting vector quantization based on intraband and interband correlation. Here the whole distortion of the vector is predicted by the part distortion of the lower frequency coefficient components of vector, resulting in a reduction of 76 percent of complexity.

The PCR products were examined by agarose gel electrophoresis. The target gene fragments were purified by gel extraction kit and ligated to cloning vector pMD18-T. The recombinant vectors were transformed into host strain E. coli K802 by lithium chloride method, screened and identified with PCR and restrictive enzymatic digestion. Their sequences were confirmed by DNA sequencing.(2) sTWEAK1 gene was subcloned into expression vector pProEx HTb and transformed into E. coli BL21. sTWEAK2 gene was subcloned into expression vector pMAL-C2x and transformed into E. coli TB1. The recombinant vectors were screened and identified with PCR and restrictive enzymatic digestion. The recombinant fusion proteins were induced to express with IPTG, detected by coomassie brilliant blue-stained SDS-polyacrylamide gel electrophoresis , and confirmed by Western blot analysis.(3) The sTWEAK1 fusion protein was purified with Ni-NTA Spin Kit.(4) The biological activity was assayed on transformed and tumor cells by microplate photometer after crystal violet or sulfur rodamine B staining.(5) The contents of IL-8 in the supernatant of 1990 cell cultures were determined by ELISA.(6) The morphological changes of the sensitive cells were observed by light and transmission electron microscopies.(7) The cell cycle and apoptotic rate were assayed by flow cytometry in 1990 and M85 cells.(8) The effect of fusion proteins on induction of NF-κB in 1990 and LOVO cells was detected with Dual-Luciferase Reporter Assay system.(9) The TWEAK gene was subcloned into Adeno-X Viral DNA with pShuttle vector and transfected into HEK293 cells by lipofectamine method.
(1)本研究用RT-PCR方法,从人组织细胞总RNA中扩增可溶性TWEAK胞外区(sTWEAK1和sTWEAK2)的cDNA序列及全长编码序列,用琼脂糖凝胶电泳分析PCR产物,胶回收目的基因片段,连接到pMD18-T克隆载体中,转化大肠杆菌K802,PCR和酶切筛选阳性克隆,全自动DNA测序验证序列;(2)sTWEAK1和sTWEAK2分别亚克隆到pProEx HTb和pMAL-C2x表达载体中,分别转化大肠杆菌BL21和TB1,PCR筛选和酶切鉴定,阳性克隆用IPTG诱导表达,表达产物用SDS-PAGE分析和Western blot验证融合蛋白;(3)用NTA-Ni Spin试剂盒初步分离纯化sTWEAK1融合蛋白;(4)用体外培养的肿瘤细胞和正常对表达产物进行活性检测,贴壁细胞用结晶紫染色法,悬浮细胞用磺酰罗丹明B染色法,酶标仪检测OD值;(5)敏感细胞用ELISA法检测细胞培养上清中IL-8的含量;(6)用光镜和电镜观察敏感细胞死亡和细胞凋亡情况;(7)用流式细胞仪分析表达产物对敏感细胞凋亡率和细胞周期的影响;(8)用双荧光素酶报告基因检测法,测定表达产物对敏感细胞NF-κB的影响;(9)用pShuttle穿梭质粒将TWEAK重组到腺病毒载体上,用脂质体转染法转染HEK293细胞,PCR鉴定重组质粒。

The total proteins from TGIF transfectant and vector control cells were separated by 2D electrophorosis. 760±41 and 680±38 spots were detected in TGIF transfectant and vector control cells respectively. Locus repeat analysis of protein spots showed that the mean deviation in IEF direction in TGIF transfectant and vector control cells were 0.864±0.123 mm and 0.843±0.115 mm respectively whereas the mean deviation in SDS-PAGE direction in these two cells were 0.812±0.109 mm and 1. 125±0.123 mm respectively. So the well-resolved and reproducible 2DE patterns from TGIF transfectant and vector control cells were established.

The second part was to test the appearance, ingredient analysis, surface potential, thermal conduction properties, and magnetism of the nano composite fluid prepared from the parameters. The experiment found that unoxidized particles, Ag, Cu, Fe, and Ni appeared round. If combined with oxygen in water as oxides, the nano particles would grow toward certain directions. In copper/iron nano composite fluid, FeO appeared cubic, Cu2O appeared coniferous. In silver/iron nano composite fluid, FeO was polygonal. In dielectric potential detection, the nano composite fluid was likely to aggregate and deposit, except for silver/iron set. In other sets, the pH of silver/cobalt nano composite fluid was 7, surface potential was 21.21mV; the pH for silver/nickel nano composite fluid was 6.5, surface potential was 21.04mV; the pH for copper/iron nano composite fluid was 7, surface potential was -30mV. The fluid particles of the three sets could all maintain suspension of 2 weeks or more. For thermal conduction, the silver/nickel, nano composite fluid showed the best thermal conduction. Under temperature of 30℃ fluid weight conduction of 0.4%, the thermal conduction increase was 26%. For magnetic detection, except for silver/iron nano composite fluid, the nano particles of other three sets were paramagnetic, and all four sets were soft magnetic nano composite materials.
第二部分。将由较佳制备参数所产出的奈米复合流体,进行形貌外观、成分分析、表面电位及热传导性质实验与磁性检测;在奈米颗粒的形貌部分,经实验发现Ag、Cu、Fe、Ni等未氧化的颗粒皆呈现近似圆形,而若与水中的氧结合形成氧化物,奈米颗粒则会朝特定方向成长,在铜/铁奈米复合流体中FeO为四方体结构、Cu2O的颗粒是针叶状,在银/铁奈米复合流体中FeO则为多边形结构;而介面电位检测方面,结果显示经本制程所产出的奈米复合流体除银/铁奈米复合流体这一组较容易聚集沉淀外,其他三组中银/钴奈米复合流体的pH值为7,表面电位为21.21mV,银/镍奈米复合流体的pH值6.5表面电位为21.04mV而铜/铁奈米复合流体的pH值为7时表面电位约在-30mV,且此三组流体颗粒皆能维持悬浮性2周以上,悬浮性较佳;热传导实验部分,四组奈米复合流体中以银/镍奈米复合流体在增进热传上效果最佳,在温度30℃及流体重量浓度0.4﹪条件下热传导系数增进达26﹪;磁检测方面,除了银/铁奈米复合流体外,其他三组奈米颗粒皆属顺磁性,且这四组奈米复合材料都是属软磁性。

Through detailed analyzing some influencing factiors of sand liquefaction, seven parameters were selected as assessment indexes. They are earthquake magnitude, peak ground surface acceleration, standard penetration value, specific penetration resistance, relative compaction, average particle diameter, and water table. The extension assessment model has been established that was applied to assessment of sand liquefaction potential. The model had been trained by the historical data and has been assessed in virtue of sand liquefaction from the historical data by simulating, and got a excellent result. After introduction of the support vector machine theory, this paper established the assessment model of sand liquefaction based on the support vector machine, and assessed in sand liquefaction from virtue data of some fields in home and overseas with the tool Libsvm, which was programmed based on the support vector machine theory. And got a good coherency between the assessement results and the actual liquefaction.
在详细分析影响砂土液化因素的基础上,选取了震级M、地面最大加速度g_、标准贯入击数N_(63.5)、比贯入阻力P_s、相对密实度D_r、平均粒径D_(50)、地下水位d_w等7个参数作为评价指标,建立了砂土液化势的可拓评价模型,并对历史数据进行仿真评价,取得了较好的应用效果;在对支持向量机的理论进行介绍后,本文建立了基于支持向量机的砂土液化势评价模型,并利用在基于支持向量机理论的Libsvm软件,对若干国内外场地的现场实测数据进行了评价,评价结果与现场表现基本上一致。
