YAC is now prevailingly used as the first cloning vector for human genome analysis, but one problem that is not yet solved is the insert DNA fragment is not easily isolated from yeast chromosomes and vectors. Na-K-Exchanging ATPase is a universal cell membrane protein in higher eukaryotic cells which mediates the anti-concentration gradient exchange of intracellular Na+ for extracellular K+. It plays essential metabolic roles such as maintaining sodium and potassium ion gradients across the plasma membrane. The targeted sequences were amplified by PCR to determine the genome structure of the hithertofore unsequenced portion of the alpha 1 subunit of the human Na-KExchanging ATPase gene which encodes 80-130 aa of its extracellular domain using two different templates, human genomic DNA and a human muscle cDNA library. The PCR products were analyzed by restriction endonuclease digestion and then cloned into a plasmid vector for chemiluminescence sequencing and further analysis.

Na-K-Exchanging ATPase是一种普遍存在于高等生物体内的细胞膜蛋白，主要参与介导K+和Na+在细胞内外之间的逆浓度梯度的转运，并维持一定细胞内外的离子梯度，我们采用聚合酶链式反应方法分别以人基因组DNA及cDNA文库为模板对人Na-K-Exchanging ATPaseα1亚单位基因胞外区约80-130位氨基酸编码序列进行扩增，限制性酶切分析扩增产物，并进行荧光测序，对测序结果进行同源性分析及剪接位点的搜索并对得到的核苷酸序列进行进一步的分析，发现人基因组DNA和cDNA经过扩增后分别得到833bp和195bp两种不同大小的片段Fg，Fc。

In this study,one pair of specific primer for mature chicken interleukin-18(ChIL-18) cDNA was designed and synthesized according to the previously published gene sequence of ChIL-18.The full length cDNA gene of ChIL-18 encoding mature active protein was amplified from LPS–stimulated MDCC-MSB1 cells by Reverse Transcription-Polymerase Chain Reaction.Then it was cloned into pMD18-T vector. Sequencing analysis showed that the nucleotide sequence of this ChIL-18 mature protein gene was 5l0bp including the stop coden and the same as the published ChIL-18 cDNA sequence by Schneider K.

本研究根据已发表的ChIL-18成熟蛋白(mature ChIL-18，mChIL-18)的cDNA基因序列，设计一对特异性引物，应用反转录-聚合酶链式反应(reverse transcription-polymerase chain reaction，RT-PCR)技术从脂多糖刺激10小时活化的马立克氏病成淋巴细胞样细胞系MDCC-MSB1的cDNA中扩增出编码鸡IL-18成熟活性蛋白的全长基因，对扩增片段进行T-A克隆，经PCR、酶切鉴定及测序验证，成功获得ChIL-18成熟蛋白完整基因的克隆。

In order to improve the inspection technologies of satellite interference, 5 kinds of technologies of satellite interference and integrated judgement-spectrum analysis, helix inspection, PID, S/N and signal vector were put forward. The automatic inspection system of interference of satellite TV has been built according to these inspected technologies and technology of network. The system adopts comparison of spectrum template, calibration of beaconing, mode identification, telemetering pick-up technology and improve the inspection technology of satellite interference. The alarm limits haw been defined through a lot of emulational tests, so that the system become feasible. The system has a good effect and makes up the blank of inspection technologies of interference in our country to some extent.
