In this paper, an efficient and easy biological screening method based on the sensitivity to lovastatin of Neurospora crassa was built to deal with the mutant strain rudely by UV mutation. A high-level strain was screened in the shaking flask culture incorporating with TLC and HPLC. A comparison between this two methods also was originally discussed while identifying dyeing using bromophenol blue as the better treatment of TLC, and offering a kind of simple, practical, fast and economical determination qualitatively and quantitatively.
本文有鉴于此,通过紫外诱变处理,并基于粗糙脉孢酶对洛伐他汀的生物敏感性,对诱变菌株进行了初筛;同时,采用摇瓶培养,结合TLC和HPLC的分析方法,复筛出一株高产菌株;对复筛过程两种分析方法分别进行了初步探讨,比较其优劣,确定TLC法经溴酚兰显色剂显色处理为较佳方式;从而,建立了一套简单、实用、快速、经济且有效的洛伐他汀生产菌诱变筛选方法。

A bacterial strain ADZ1 that demonstrated higher efficient in the aerobic denitrification was isolated from the sewage of urban rivers on the selective medium, Bromothymol Blue, after enriched in the shaking flasks. However, it is not known how orthodontic force influences the function of cementoblast and what is the relationship between the function change of cementoblast with root resorption. So we studied the effect of mechanical stimuli on root resoption by examining the expression OPG and RANKL and exploring the related signal transduction pathway in cementoblast. Methods: Cementoblasts OCCM30 were cultured in DMEM with 10% FBS for 48 hours and starved in DMEM with no FBS for 24 hours, then subjected to mechanical strain by four-point bending system with tension and compression stress at 2000μstrain at 0.5Hz frequency. The flow cytometry was used to examine the cell cycle and proliferation activity after 3h, 6h, 12h, 24h loading respectively. OPG and RANKL mRNA were analysed with real time quantitive RT-PCR after the same loading period as FCM.

The structure and stability of all possible isomers of C50X(X=SiH2, PH, S) were investigated by ab initio HF/3-21G calculations. The calculation results indicate that the preferred position for cycloaddition of SiH2, PH, and S on C50 is consistent, i.e. on the C3-C4 and C4-C4 sites of C50, and the resultant most stable cycloaddition isomers are [5,6]-closed and [5,5]-closed structures. The main driving force governing the stability of the C50X(X=SiH2, PH, S) isomers is the strain, or the conjugative effect, or the combination of strain and conjugative effect, depending on the addition positions and the types of the attacked C-C bonds.
