To evaluate the efficacy of the recombinant fowlpox virus vaccine against the heterotype IBV, the chickens were challenged with the homotype IBV LX4 strain and the heterotype IBV LTJ95I strain after three weeks post vaccination. Antibodies against IBV were detected in vaccinated chickens one week post inoculation. The CD4+ T-lymphocytes in the peripheral blood increased rapidly in all groups challenged with IBV, except for the vaccinated group challenged by heterotype strain and the low level of CD4+ T-lymphocytes remained until end of the experiment. In all the groups, a high level of CD8+ T-lymphocytes only was observed in the vaccinated group after challenging with IBVhomotype strain. The morbidity and the mortality of this group were 21.43 % and 0 %, respectively, which showed significant difference with other groups. In addition, the lesions of chickens and virus shedding were less in the vaccinated group challenged by IBV homotype strain comparing with other groups, but there was no difference for the average body weight of chickens in all groups.

For further understanding the effect of the strain on K-bearing silicate mineral, an experiment with KO2 strain cultured in nitrogen-containing and nitrogen-free medium was conducted. Another result was that there was higher level of Carbonic Anhydrase activity, which revealed that some exo-protein or enzyme produced by the bacterial had postive impact on the process of releasing potassium ion. We carried out the bacterial fermentation for larger scale production of the bacterial secretion, which was used to sperate, and identify the small molecules related to the mineral-bacterial interaction. After the analysis, it was found that the strain can produce many kinds of small molecules, such as 2-Hydroxybenzoic acid, 4-Hydroxyphenylacetic acid, methyl-4-hydroxybenzoate, etc.

The effects of strain rate on plastic formation and dynamic recrystallization of 7050 aluminum alloy were discussed. The results show that the deformation of the specimen is inhomogeneous, and the effective strain in the center of the specimen is maximum. The deformation inhomogeneity increases with the increase of strain rate. The effective stress of specimen is inhomogeneous. As the strain rate increases, the minimum stress value moves from the drum-shaped region to the heart of specimen. The standard deviation of dynamic recrystallization grain size decreases with the increase of strain rate.

But for theouter cylinder,the former is much smaller than the later at the same direction.3 Forboth the inner shell and the outer shell,hoop strains are primarily due to themembrane deformation.The amplitude of the circumferential flexural strain isconsiderably small.4 Peak strains in the vicinity of the connection portion of theend caps and the shells are very high.This is important to assess the safety of thecontainer in case of repeated use or tests.5 The applying load to the end cap isdifferent from that to the inner shell plate, but peak stains and nodal displacementsof these two structural parts are nearly the same.It can be inferred that the dynamicresponse of the end cap is greatly affected by the structural vibration excited by theimpulsive loading.

One pair of primers that amplified the gB gene of pseudorabies viruswas designed and synthesized.PCR technique detecting the DNA of PRV was established after selecting the best reaction conditions.This technique was applied to specifically amplify the 281 bp DNA fragment of the PRV strains including Fa,Fb,Bartha,BJ,GD,V2F4,S,S3,SR,Buk,Shope,Norden,Mink Ⅲ,HB,F8,F9 and F12 in cultured samples.The negative results were achieved from Vero cells,swine vesicular disease virus,hog cholera virus,Japanese encephalitis virus,porcine reproductive and respiratory syndrome virus,porcine parvovirus,foot and mouth disease virus F29 strain,O3I3 strain,T509 strain and O Ⅱ MF249 strain.The results of sequencing showed that the PCR method was of specificity.The sensitivity of PCR reached 15.8 pg of PRV Fa strain DNA.The tissue samples obtained during 1994 and 2000 were detected,and the results showed that the sensitivity of PCR was more sensitive than virus isolation and the Sandwich ELISA.The PCR was applied to detect 191 tissue samples from 31 pig farms obtained from Guangdong,Fujian,Hainan Provinces during 1999 and 2000,50 samples(26.2%)were positive and 22 pig farms(71%)were positive.

The main research tasks and results are as follows:(1) Through comparing the stress intensity factor of compact tension specimen calculated by finite element software with the theoretical value, the feasibility of calculating fracture parameters of crack tip in ABAQUS software was verified.(2) The relationship between the elastic modulus of the material and research scale was studied based on deriving the cohesive stress theory and other relevant theories. And the elastic modulus of the material on meso-scale was determined.(3) Global model and sub-model of the finite element were built in th...e macro-scale, and stress-strain field nearby the crack tip was analyzed, and the results show that the detailed stress-strain in the crack tip could be obtained by using the sub-model technique.(4) In meso-scale, the crack growth model was also established by assuming the crack propagating along the grain boundary, and the detailed stress-strain field in the crack tip was obtained.(5) The effect of crack length on Von Mises stress, and maximum principal stress and strain was analyzed in meso-scale when equivalent KI equals 30MPa.m1/2. And the results show that except for crack tip area, the stress-strain distribution in whole specimen is not affected.
