Based on the cloning FeSOD gene of the HZNH, the recombinant prokaryotic expression vector, PQESOa/FeSOD, was constructed and digested with restriction endonuclease BamH I and Pst I to check its construction. The*results showed that the target proteins held a 37% portion in whole bacterial proteins and consisted of two parts, the soluble proteins and inclusion bodies. The soluble proteins in the aqueous layer, checked by means of activities of FeSOD enzymes and analyzed by means of activities of isozymogram from PAGE, demonstrated the induced expression proteins had the active nature of FeSOD enzymes.

In this study, we isolate, purify and characterize endo-β-glucanase by Ascomycete Xylaria regalis. The cellulose-containing medium is used to induce the production of the cellulose-degrading enzymes. When these enzymes are produced at maximum level, we harvest and filtrate the culture medium. The filtrate is purified by anion exchanger column and hydrophobic interaction column (phenyl-650M). The purification fold of endo-β- glucanase increases 4.3 times, the specific activity approaches 3.7 unit/mg, and the recovery is 27.6%, respectively.

The results show that the optimal hydrolysis conditions of these three enzymes were:50℃,pH 8.5, substrate concentration 1:3,enzyme dosage 1200U/g,time 4h for Alcalase; 45℃,pH 8.1, substrate concentration 1:3, enzyme dosage 1200U/g, time 4h for Trypsin;50℃, pH5.5, substrate concentration 1:3, enzyme dosage 1400U/g, time 6h for Bromelin, respectively.
