This research is the case study of Furniture Library. It is a research of inner space planning of the Furniture Bibliotheca HDG. Besides doing research on subject related documentations, this paper also designs questionnaires in regards of the readers demographic, library space allocation and business management of Furniture Bibliotheca HDG. Based on the analysis and result of the questionnaires, the paper will provide suggestions and serve as a reference for Furniture Bibliotheca HDG to enhance their service quality.

On the basis of total analysis and summarization on basic knowledge, principle and expert experience of injection mould feed system as well as further research on the computer-aided design of injection mould feed system, according to the geometrical model of the plastic, applying technologies including feature-based modeling and identifying, RBR, CBR, oriented-object and database technology VC++ 6.0, SQL Server 2000, Object ARX, the paper conceives general structure and primary function module of the intelligent design of injection mould feed system, develops user-systemic interface, constructs knowledge rule library and case library of injection mould feed system, researches denotative method of knowledge rule library and case library, designs the ratiocinative mechanism of intelligent system design, and realize the intelligent design of injection mould feed system partly.

本文在对注塑模具浇注系统设计的基本知识与原则及专家经验全面分析总结，以及对计算机辅助浇注系统设计方法深入研究的基础上，以塑件几何模型为依据，应用基于特征的建模与识别技术、RBR和CBR推理技术、面向对象和数据库技术，以及VC++6.0、SOL Server 2000和Object ARX二次开发工具，在AutoCAD 2002平台上，构建了注塑模具浇注系统智能化设计系统的总体结构及主要功能模块，开发了系统的用户界面，建立了浇注系统设计的知识与规则库和事例库，研究了设计知识与规则和事例的表示方法，设计了智能化设计系统的推理机制，部分实现了注塑模具浇注系统的智能化设计。

Library circulation record analysis helps to understand users' needs and satisfaction. This research is the first one that uses graphic method to analyze Library circulation records baseing on Complex Network. The researcher built Library circulation network from Library circulation records through the relationship between users and that between users and books. The research attempts the following analyses:(1) Properties of Library circulation network;(2) The phenomenons and trend of Library circulation network;(3) Motif inside Library circulation network.

The researcher provides suggestions for further development in the following eight respects:(1) promote the union catalog of ISBNnet to libraries with limited human resources so as to improve the administration efficiency;(2) integrate the personalized service functions into the library acquisition system in order to provide customized alert services for new publication information;(3) embrace RSS into the library acquisition system to make it easy for library users to request real-time purchase recommendation;(4) add the mechanism for group learning to the library acquisition system to facilitate the sharing of professional knowledge and interaction;(5) appeal to the Central Trust of China in Taiwan to amend the regulation on order arrival rate in the Inter-Entity Supply Contract;(6) advocate the union catalog to include bibliographic records of publications in languages other than Chinese;(7) encourage the library to launch more practices of e-services to enhance administrative efficacy;(8) improve the quality of other library services by employing the action research method in library practices.

The results of a 1987 survey research are reported. The most current course syllabi on the Web are also examined to analyze and compare the characteristics of the content of the research methods courses in library schools.

All index proved that the P5776 cDNA library constructed in this research had high quanlity and could be used for further ESTs sequencing and analyzing. The amplified library of P5776 was screened on white-blue plates, and more than 5,000 white-clones were selected to culture in 96-well plates, then the high quanlity plasmids of white-clones were extracted by traditioal alkaline-lysis method using the Vitagene?96-easy plasmid extract kit. Through PCR primered by T3( the vector has T3 RNA polymerase promoter ), the target fragments of inserts were get. Then the inserts were taged by four different fluorescence-dye, and fractionated by capillary in ABI Prism? sequencer. More than 5,000 5′-ESTs had been sequenced and through rough selecting, 4,747 ESTs were remained for further research. Among the remained ESTs, more than 90% have longer lenth than 500bp, and there are less than 5% unable-read bases in every sequence.

将扩增好的P5776 cDNA文库铺布蓝白斑筛选平板，挑取其中的白斑经活化培养后，运用Vitagene 96-easy质粒DNA制备试剂盒以碱裂解法共提取出58 板(96孔板)，即5，000 余个重组子克隆的高质量质粒DNA；提取好的质粒以T3(连接载体上具有该引物RNA聚合酶启动子)为引物经PCR(Polymerase Chain Reaction，聚合酶链式反应)扩增出重组子插入片段，标记上荧光染料后在ABI Prism 3100 测序仪上经毛细管电泳完成了5，000 余条5′端EST 序列的测定工作，初步筛选出4，747 条质量较好的EST序列，经筛选得到的EST中90%以上具有大于500bp 的可读序列，每一EST序列中不可读碱基数小于5%。
