The different fishes which are employed in the present studies are wild-type salmon, cultured salmon of freshwater and seawater, sea perch and fat greenling. The complete CT gene sequences of salmons are obtained by PCR amplification. The partial CT sequences of sea perch and fat greenling are obtained by in vitro cloning PCR method. Alignment of obtained CT sequences with other fish CT shows that CT appears to be well conserved among the same family. And the relative in taxonomy is far away, the similarity of different fish CTs is low. On the contrary, the closer the relative in taxonomy is, the higher the similarity of different fish CTs is. The sCT is expressed in pGEX-4T-X by recombinant form . We also succeed in the research of sCT expression alone by expression PCR. In addition, sCT antiserum is obtained using GST-sCT as antigen, and the high titer is tested by double immunodiffusion. In the rat bioassay, administration of 50 μg recombinant protein evoked significant hypocalcemia at 1 h after the data are analyzed by t-test.
本文用PCR方法克隆了野生鲑鱼、养殖鲑鱼的降钙素基因,并应用体外克隆PCR的方法首次克隆出鲈鱼、六线鱼降钙素的部分基因序列,通过对克隆的降钙素序列的比较研究,结果显示同一科的鱼降钙素序列保守性较高,同时,根据降钙素的部分氨基酸序列进行了降钙素相似性的比较研究,结果显示在分类学上,分类地位较远的鱼,其降钙素相似性较低,分类地位越接近的鱼,其降钙素相似性越高;我们利用谷胱甘肽S-转移酶(Glutathions S-transferase,GST)融合表达载体pGEX-4T-X对克隆的鲑鱼降钙素基因进行了融合表达研究,应用表达PCR的方法对降钙素基因的独立表达进行了初步的研究探索,并将纯化的融合蛋白作为抗原,获得了高效价的兔抗鲑鱼降钙素免疫血清;生物活性研究表明,大肠杆菌表达的融合蛋白具有显著的降血钙作用。

This paper studies depression in the hot cathode glow discharge PCVD method of diamond films influence in the process, mainly from two aspects of research: a study of the process of discharge pressure of the impact of that pressure on the technology, the impact of state discharge , as well as pressure and discharge voltage of the relationship between the two, and experiment with different pressure, relative to the preparation of samples, and using Raman instrument and SEM analysis of the quality of the film, by comparing the different types of gas pressure on the quality of diamond films, summed up the most good gas pressure parameters of a high-quality diamond films.
本文研究气压在热阴极辉光放电PCVD方法制备金刚石膜过程中的影响,主要是从两个方面进行研究:一,研究制备过程中气压对放电的影响,得出气压对工艺,放电状态的影响,以及气压和放电电压的关系;二,采用不同气压进行实验,制备出相对应的样品,并用Raman和SEM等仪器对膜进行质量分析,通过对比不同气体压强对金刚石膜质量的影响,总结最佳气体压强参数,制备出高品质金刚石膜。

At first, this paper conducts in depth research on classification method, compaction propeties, granulometric composition and crushing of stone. On the basis of clarifying engineering characteristics of material of rock-filling embankment, requirements of ground treatment have been put forword in this paper. Then combined with the site test and engineering practice in SHUINAN Road and LANLIN Road, through the synthetical analysis of factors of influencing densification effect for the maximum grain size of filling material, lay-down thickness, granulometric composition, densification machinery and densification frequency etc., this paper studies key technical problems about construction technologies for spreading and leveling of filling material, choosing densification machinery, densification technology and slope construction systematically. Finally, construction technologies of filling lime-stone and sand-stone roadbed have been proposed in this paper ,which are significant for quality guarantee of highway engineering.
本文首先从石料的分类方法、压实特性、粒径组成以及破碎性等方面进行研究,在明确填石路基填料的工程特性基础上,进而提出了填石路基的地基处理要求,然后结合广西水南路和甘肃兰临路填石路基试验路段上的现场研究,通过对填石路基填料最大粒径、松铺厚度、粒径组成,压实机械以及碾压遍数等影响压实效果因素的综合分析,系统研究了填石路基填料的摊铺与整平技术、压实机械选型技术、压实工艺以及边坡施工技术等施工工艺中的关键问题,最后提出石灰岩类和砂岩类填石路基的施工工艺要求。

This paper presents a review of studies of interference absorbing materials,which come in three basic types:normal interference absorber,Jauman absorber and FSS absorber. And we particularize the structures,applications and methods of analysis and design of each of these types. Results show that the FSS absorber provides a better absorption ability and is more practicable,and that active multi-layer FSS absorber represents a new direction of interference absorbing material research.
