Object: The project is on the basis of chronic alcoholic myopathy model of rat, to dynamically observe the change of NF-κB and MMP-2, MMP-9 in muscle cell, observe the function in the mechanism of alcohol myopathy, move forward a single step to explain the pathogenesy of this disease from gene and protein leve. We also provide a helpful platform for other metabolic myopathy. Methods:(1) 80 health, male, Sprague-Dawley rats(weight 180~200g), were chosen to sep up the CAM rat myopathy model ,to observe the rat bodyweight, the change of praxiology and the patho-change of muscle tissue.
目的:本实验旨在建立大鼠慢性酒精中毒性肌病模型的基础上,动态观察NF-κB及MMP-2、MMP-9在酒精中毒性肌病发展中的表达、变化情况,探讨其在慢性酒精中毒性肌病发病过程中的作用,从基因和蛋白水平进一步阐述该病发病机制,为慢性酒精中毒性肌病的临床防治提供理论依据,同时对于其他代谢性肌病的研究也提供一个平台。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.
研究方法:(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC,细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构;(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定;(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC,并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况,根据各个细胞克隆受Dox调控表达的程度,选择低背景、高诱导表达AT2R的细胞系,作为双重稳定MSC细胞系;蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况;(4)建立大鼠颈动脉球囊损伤动物模型,将双重稳定MSC在术中导入血管,分别于14 d、28 d进行病理切片,检测可调控AT2R对新生内膜增生的影响;采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变,TUNEL法检测血管组织中细胞凋亡的变化情况。

Rresults from those studies can be summariazed as follow:(1) A preliminary study no correlationship between the UV-VIS reflection spectrum of tomato fruit surface and the lycopene amount of tomato fruit was undertaken. Based on observation from this study, a damage-free and in-situ detection technique of lycopene amount from tomato was developed with UV-VIS reflection spectroscopy;(2) The geometrical isomers of lycopene were separated successfully on C30-HPLC-PDA-ELSD. The absorption coefficients of Z-isomers were consequently dedermined. The geometrical isomer composition of lycopene could therefore be assessed. Those efforts formed a solid base to identify the natural product from synthetic compound of lycopene;(3) Lycopene resource from the fruit of Autumn oliver was proved to be available for industrial application. An extraction methos of lycopene from the fruit of Autumn oliver by supercritical carbon dioxide was developed at laboratory scale;(4) A method to extract lycopene by supercritical liquid with improved efficiency was developed; Data from this investigation suggested that a proper raw material pre-extraction process was important for a better extraction efficiency;(5) Variation in the geometrical isomer composition of lycopene during its metabolish in rat was assessed. A large amount of Z-isomers were observed. This variation took place in serum;(6) The absorption rate and accumulation in serum of natural lycopene in rat were assessed;(7) Ability to quench singlet oxygen by different geometrical isomers of lycopene was compared;(8) It was detected that whether natural lycopene had functions to regule blood-lipids and LDL antioxidation;(9) A C31 degradation piece of lycopene was found in the prostate gland of rat.

Antinociception and VGSC-modulation of BmK IT2 and BmK AS—1. The results from nociceptive flexion reflex and plantar radiation heat showed: BmK IT2 (a depressant insect-selective neurotoixn) and BmK AS-1 (a new-type β-1ike neurotoxin) have significant antinociceptive effects on rat peripheral nerve system. The antinociceptive effects of BmK IT2 on the carrageenan-inflamed rats were significantly stronger than those on the normal rats. Whole-cell patch-clamping showed: Both TTX-S and TTX-R Na〓 currents induced from rat DRG neurons were significantly inhibited by BmK IT2 and BmK AS—1, and BmK IT2 was more sensitive to TTX-R than TTX-S component. All the results suggested: the peripheral antinociception of BmK IT2 and BmK AS—1 might be attributed to the inhibitory effects of them on the TTX-S and TTX-R Na〓 channel located on the peripheral ends of rat DRG neurons with small diameter.

The mature region DNA for Myostatin of Xiaoshan chicken has the identity of 99%, 97%, 93%, 92%, 92%, 87%, 86%, 86%, 86%, 86%, 85%, 85%, 85%, 84%, 84%, 83% and 74% compared with that of chicken, turkey, pigeon, goose. Fan duck, monkey, human, horse, pig, Jinhua pig, house mouse, rat, sheep, hare, goat, cow and zebrafish respectively. The mature region DNA for Myostatin of Fan duck has the homology of 91%, 92%, 92%, 90%, 95%, 84%, 84%, 85%, 85%, 85%, 83%, 81%, 81%, 83%, 81%, 82% and 73% compared with that of chicken, Xiaoshan chicken, turkey, pigeon, goose, monkey, human, horse, pig, Jinhua pig, house mouse, rat, sheep, hare, goat, cow and zebrafish respectively.
