Methods A nested RT-PCR was used for detecting one fragment of SARS-CoV RNA in oropharyngeal swabs from 3 SARS probable patients ,4 SARS suspect patients and other 27 patients with fever in Hangzhou, and the nested RT-PCR product from one SARS probable patient was sequenced.Meanwhile in these 3 SARS probable patients, other three RT-PCR methods, including a hemi -nested RT-PCR targeted for another fragment of SARS-CoV RNA, a real-time RT -PCR and a modified nested real-time RT-PCR, were employed to detect SARS-Co V RNA.Results Two positives were found in the 3 SARS pro bable patients, and none positive in 4 SARS suspect patients and other 27 patien ts with fever, using the nested RT-PCR.

方法对2003年杭州市3例严重急性呼吸综合征临床确诊患者、4例疑似和27名医学观察者的咽拭子标本用巢式实时RT-PCR检测SARS-CoV核酸，对阳性扩增产物进行核酸序列测定，同时对3例患者的标本应用半巢式RT-PCR检测另一位点SARS-CoV核酸片段，并应用常规实时RT-PCR技术及改良实时RT-PCR技术进行检测。

Meanwhile, it could be seen that there does not exist a confidence interval with width less than 0.5 because of the property of Poisson distribution. Based on these conditions, the author mainly carried out research into two aspects of this problem as follows. Firstly, by numerical and theoretical analysis, the author compares some existent confidence intervals, for example,"exact" confidence interval, Wald confidence interval and Bayesian confidence interval, and finds some deficiencies points of the confidence intervals, whose modification version has been proposed .Also, several better confidence intervals such asare also presented .Secondly, for given confidence coefficient and interval width, the author constructs a class of asymptotical two-stage interval estimate procedures. At the same time, under varies restriction of confidence coefflcientent interval width, the optional sample size of the first stage has been computed by numerical computation. The numerical computation shows that the method considered in this dissertation have good properties and applied value.

Therefore,tapping potencial of remaining oil to improving oil recovery,the recognition of reservoir intraformational heterogeneity in channel sandbodies is proposed and the main targets of reservoir IOR in channel sandbodies of mature oilfields are pointed out as follows:①shutting off strongly-waterflooded intervals to control ineffective circulation of injected water;②strengthening water washing in medium-waterflooded intervals in order to achieve the aimed waterflood efficiency and even to exceed it;③enhancing injection-production consistency and injector-producer ratio of non-waterflooded or weakly-waterflooded intervals for the propose of realizing effective displacement and imporving conforma nce.
