Additionally, the growth of the pathogen was obviously affected by cultural media. The morphological characters such as colony color and diameter, and production of fruit body of the fungus growing on PDA, 10% V8, apple leaves dextrose agar and apple leaves extraction dextrose agar media were different. The fungus formed colonies of about 8mm in diameter and did not produce fruit bodies and aerial hyphae in 1 month incubation on PDA. However it formed the similar size colonies as on PDA and produced fruit bodies and aerial hyphae on 10% V8 and LDA media in the same incubation period. Very small colonies (2mm in diameter) and fruit bodies were found on LEDA media in the same incubation condition. These results indicated that successful isolation of M. coronaria from apple leaves depended on suitable isolation method and cultural media as well as fresh samples.

Rresults from those studies can be summariazed as follow:(1) A preliminary study no correlationship between the UV-VIS reflection spectrum of tomato fruit surface and the lycopene amount of tomato fruit was undertaken. Based on observation from this study, a damage-free and in-situ detection technique of lycopene amount from tomato was developed with UV-VIS reflection spectroscopy;(2) The geometrical isomers of lycopene were separated successfully on C30-HPLC-PDA-ELSD. The absorption coefficients of Z-isomers were consequently dedermined. The geometrical isomer composition of lycopene could therefore be assessed. Those efforts formed a solid base to identify the natural product from synthetic compound of lycopene;(3) Lycopene resource from the fruit of Autumn oliver was proved to be available for industrial application. An extraction methos of lycopene from the fruit of Autumn oliver by supercritical carbon dioxide was developed at laboratory scale;(4) A method to extract lycopene by supercritical liquid with improved efficiency was developed; Data from this investigation suggested that a proper raw material pre-extraction process was important for a better extraction efficiency;(5) Variation in the geometrical isomer composition of lycopene during its metabolish in rat was assessed. A large amount of Z-isomers were observed. This variation took place in serum;(6) The absorption rate and accumulation in serum of natural lycopene in rat were assessed;(7) Ability to quench singlet oxygen by different geometrical isomers of lycopene was compared;(8) It was detected that whether natural lycopene had functions to regule blood-lipids and LDL antioxidation;(9) A C31 degradation piece of lycopene was found in the prostate gland of rat.

The ethanol content in fruit flesh of'Starking'apple, 'Yar'pear and'Shingo'pear increased in early storage with ethanol-applied concentration increased and decreased later. At the end of the storage for 5-6months, there was no difference in ethanol concentration in fruit flesh in all treated apple or'Yar'pear treated with 2 and 4ml/kg fruit of ethanol and control, but that in 6ml/kg fruit of ethanol treated'Yar' pear was higher than that in control.

MCP could effectively delay the softening of climacteric fruit, such as Starking and Fuji apples and Feicheng peach, but as different fruits had different softening mechanism, the mechanism of 1-MCP inhibiting different fruit softening was different: in Starking apples, the key enzyme to the softening is endo-and exo-PG, while during Feicheng peach fruit softening, the key enzyme was EGase before the 20th d and endo-PG after 20th d during storage at 0℃. 1-MCP inhibited fruit softening mainly by inhibiting PG activity in Starking apple while in Feicheng peach, by inhibiting the activity of EGase in the earlier storage time and endo-PG in the latter time. It was also found that LOX accelerated the initiation of the softening and was regulated by ethylene and 1-MCP in apple and peach.
