OLH or 8-bromo-cAMP were added to luteinized granulosa cells. Results indicated that, protein expression of HIF-1a, HIF-1b and VEGF had a better performance in hypoxia than in normoxic condition, but phospho-PKA, StAR and P450scc protein expression had weaker expression in the former condition than in the later one. As a result of decreasing secretion of progesterone the decreasing of phospho-PKA was suspected to have a role in the reduction of StAR and P450scc in the condition of hypoxia.

GAL-CAD achieved more than 100-fold induction in A549 cell and more than 40-fold in HepG2; expression level of the system markedly increased after addition of glycin arm, almost 10-fold enhancement in HepG2 cell. Because basic expression also increased, the induction fold by hypoxia decreased.

目的 探讨脂肪组织中是否存在缺氧预适应现象,构建cocl2致缺氧的缺氧模型。方法不同浓度的cocl2大鼠腹股沟区皮下层浸润注射,分别用免疫组织化学法和rt-pcr法检测该区脂肪hif-1α蛋白和hif-1α mrna的表达情况。结果用不同浓度的cocl2行化学缺氧后在不同时间点观察有不同程度的hif-1α蛋白和hif-1α mrna表达,其中,在浓度为20~40 mg/kg,时间为3 h时脂肪组织中hif-1α表达达高峰。结论脂肪组织存在缺氧预适应现象;cocl2的浓度选择20~40 mg/kg,时间选择3 h时缺氧预适应效果较佳。
