The hFCECs were cultured by sticking tissues piece method that the cornea were divided into the limbus and central tissue piece and digestive method,respectively.For digestive method,the digestive juice of 5mg/ml DispaseⅡ+0.25%trypsin was chosed.D/F12 with 10%fetal bovine serum and 100 U/ml penicillin and streptomycin were used in this study,the culture condition was 37℃and 5%CO2,and culture medium changed every three days2、Passage of hFCECsAfter more than 80%confluenced,cells from corneal limbus hFCECs were digested for 5-15min with different concentrations of trypsin/EDTA at 37℃,and then passaged at a ratio of 1:2.The cells were subcultured on empty plates,mouse 3T3 fibroblast feeder layer,fetal corneal stromal cell feeder l ayer or HTK feeder layer respectively.And D/F12, mouse 3T3 fibroblast conditioned medium,fetal corneal stromal cells conditioned medium or HTK conditioned medium with 10%FBS and 100 U/ml penicillin and streptomycin were used respectively as the culture medium.And the culture medium were changed every three days as well.

方法一、人胎儿角膜上皮原代和传代培养1、原代培养严格无菌操作获取人胎儿角膜片，采用组织块贴壁法、酶消化法原代培养人胎儿角膜上皮细胞。2、传代培养角膜缘部的细胞生长融合达80%以上，不同稀释浓度的胰酶/EDTA消化液消化传代分别接种于空板、小鼠3T3成纤维细胞饲养层、胎儿角膜基质细胞饲养层及HTK饲养层，培养液分别采用D/F12、小鼠3T3成纤维细胞条件培养液、胎儿角膜基质细胞条件培养液、HTK条件培养液。

RESULTS: Among 200 cases operations, 7 cases posterior capsular rupture, 2 cases rupture of the iridodialysis, 52 cases corneal endothelial edema after 1 day, 11 cases hyphema., 3 cases IOL dislocation, 2 cases incision leakage.

结果：200例术眼中，后囊膜破裂7例，虹膜根部离断2例，术后1d角膜内皮水肿52例，前房积血11例，人工晶状体偏位3例，切口渗漏2例，术后1mo，视力均＞0.2，术后6～18mo视力≥0.5的181例，占90.5%，平均散光（0.97±0.52）D。

objectiveto investigate the diagnosis and treatment of traumatic duodenal rupture.methodthe clinical data of 13 patients with traumatic duodenal rupture hospitalized from may 1991 to december 2008 were retrospectively analyzed,and the material cause was automobile accident mainly.resultsin these 13 patients,after hospitalization,6 cases were diagnosed in 3 hours,4 cases were diagnosed in 24～72 hours,and 3 cases were diagnosed after 72 hours.three cases with duodenal cleft repair,2 cases with duodenal ostomy,2 cases with duodenum and jejunum roux-en-y anastomosis,1 case with duodenal diverticular surgery,1 case with meliorative duodenal diverticular surgery,1 case with duodenal broken ends closure and anastomosis of stomach with jejunum,1 case with duodenal distal closure and anastomosis of duodenal local with jejunum,and 2 cases with resections of pancreas duodenum.after the operation,6 cases (46.2%) appeared some complications of wound infection,duodenal fistula,digestive tract hemorrhage,pancreatic fistula,abdomen infection and so on.two cases (15.4%) died from multiple system organ failure and septic shock.conclusionthe early diagnosis and correct operation style selection are the keys for duodenal rupture.

目的：探讨外伤性十二指肠破裂的诊断和治疗方法。方法：回顾分析外伤性十二指肠破裂患者13例临床资料。结果：入院后3 h内确诊6例，24～72 h内确诊4例，72 h后确诊3例。3例行十二指肠裂口修补，2例行十二指肠造瘘，2例行十二指肠空肠roux-en-y吻合，1例行十二指肠憩室化手术，1例行改良十二指肠憩室化手术，1例行十二指肠两断端关闭、胃空肠吻合，1例行十二指肠远端关闭、近端与空肠端侧吻合，2例行胰十二指肠切除术。术后6例(46.2%)出现切口感染、十二指肠瘘、消化道出血、胰瘘或腹腔感染等并发症，2例(15.4%)患者死亡，分别死于多器官系统衰竭和感染中毒性休克。结论：早期诊断与正确的术式选择是成功救治外伤性十二指肠破裂的关键。

The main research work and its results are: 1Empirical results are analysed for the relations of fault displacement and earthquake magnitude, and difference between permanent ground deformation and maxium dynamic relative displacement on the two sides of fault; 2An analytic method is proposed for resopnse of a buried pipeline due to earthquake fault movement; 3A new shell model with an equivalent boundary and relted finite element analysis method are proposed for estimating response of a buried pipeline under large fault movement; 4By the proposed method, damage performance of water supply steel pipelines with large diameter is simulated to the real cases in Kocaeli Earthquake and Ji-Ji Earthquake, and simulating results show the real failure mode of pipes is revealed; 5Effects of overlying soil and soft/hard interlayers are analyzed on ground rupture mode and degree due to earthquake fault, and some earthquake rupture phenomena in soil layers are theoretically described for the first time; 6Pipeline response characters are discussed for the case of considering effects of soil layers on ground rupture mode; 7Research results and proposed method are applied in seismic analysis of pipline acrossing fault in the Gas Tansportation Project from West to East in China.

主要工作和成果包括：1）分析了不同断层位错形式下地表破裂位错量与震级之间的经验关系，并基于集集地震近断层记录分析了断层两侧永久地面变形和地震动相对位移幅值之间的差别，为合理地考虑地震断层位错地面变形动态影响提供了依据；2）发展了断层位错地面大变形下管线反应分析模型并给出物理概念明确、简单实用的解析分析方法；3）建立了断层位错下埋地管线反应分析的等效边界壳模型有限元方法，方法中引入了以非线性弹簧形式模拟离断层较远处管段影响的等效边界，简化了计算模型并突出了近断层处管体反应；4）利用等效边界壳模型方法，对土耳其地震和集集地震中大口径埋地管线震害进行了模拟，结果表明所提方法可以更清地解释震害所表现的管体破坏特征；5）研究了覆盖土层和软硬夹层的存在对断层位错地面破裂形式和程度的影响，首次从理论上解释了某些地震地表破裂现象；6）探讨了考虑土层对断层位错地面变形影响的管体反应特征；7）本项目成果已在西气东输工程的管道跨断层抗震问题分析中得到了应用。

Experimental results demonstrate that when the tensile speed is fast, longitudinal volume electrical resistance decreases with the increasing of the stress, before the rupture the electrical resistance decreases to the most low, the moment the sample rupture, the electrical resistance increases suddenly. when the tensile speed is slow, longitudinal volume electrical resistance decreases with the increasing of the stress during the tensile prophase, before the rupture, the big surging of the electrical resistance occurres.
