To study the molecular mechanism of Larix somatic embryogenesis, a differentially expressed cDNA library of Larix somatic embryo in the period of maturation was constructed using suppression subtractive hybridization. The cDNA from the cultures at the stage of somatic embryo maturation of embryogenic cell line Y35 of L. leptolepis ×L. principis-rupprechtii was used as the tester and the cDNA from its subcultured callus was used as the driver. Eight hundreds randomly selected positive clones were sequenced, and 468 UniGenes were obtained finally.

The results showed that the optimized content of Mg~(2+) was 5mmol/L; dNTP was 200μmol/L; Primer was 0.5μmol/L; Taq polymerase was lU/50ul in the PCR system. Only one fragment of about 270bp was amplified within the genome of 12 wild species and 23 cultivars of Mallus Mill., which indicated that Ty1-copia-like retrotransposons were archaic components of apple genome. The nested insertion of near and within the reverse transcriptase gene of Ty1-copia-like retrotransposons wasn\'t found in apple genome.2 The reverse transcriptase conservative region of Ty1-copia-like retrotransposons were amplified from Gala apple using the optimized PCR system. The amplification product was isolated, cloned and sequenced. Forty-five clones containing reverse transcriptase conservative region were obtained (accession number: AY849580 -AY849592 and DQ105035-DQ105066). Cluster analysis of these sequences showed a great heterogeneity among RT domains isolated from the same genotype.

The extensive variation in chromosome number in root tip cells of three Chinese cabbage-cabbage monosomic alien addition lines with 21 chromosomes, 6-2-4, c-2-4 and d6, was obsevered after vegetative propagation multi-generations. The chromosome number varied from 17 to more than 50. The 25S rDNA signals were very complex in the three different clones.

The signals look like noncontinuous beads on the DNA fiber,which consist of multi-copy and arrange tandemly.Fiber-FISH results with 45S rDNA probe showed that an average signal length is about 3~11μm in many tandem copy sequence(measure number,n=8),so we estimated the size of each copy to be approximately 11~30kb in cotton genome.There are dual signals on each end of the mitosis metaphase chromosomes and pachytene chromosomes hybridized with telomere DNA probe.The signals also look like non-continuous beads on the DNA fiber,the length is about 1~9μm,so we estimated the size is about 4~27kb.The signals almost covered the whole mitosis metaphase chromosomes and pachytene chromosomes of G.arboretum Shixiya 1 hybridized with gDNA probe,the euchromatin zone and heterochromatin zone were identified clearly on pachytene chromosomes,and the signals also look like non-continuous beads.Two BAC clones 150-D-24 and 182-F-9 in DNA BAC library of Pima90 were selected as probe to hybridize with mitosis metaphase chromosomes,pachytene chromosomes and DNA fiber of G.raimondii.
