The change of natural aging mice's neuron in hippocampal area were studied by applying microscope and transilluminating electric microscope. Results: Comparing with the young control, the old control mice's hippocampus had less small pyramid cells, the nerve cells denaturalizing and their branches reducing in the microscope. Hippocampal nerve cells were degenemiceing, with nucleus membrance crinkling like wave shape were not clear, and chromatin condensing with high electron density, heterochromatin accruing, part cell organs inclosing nucleus, nucleus condensing in the EM.

The results showed that:(1) In COPD group the alveolar walls were broken. The average area of alveolars was 2.46 times as big as the control group, and the average diameter was 1.42 times as compared with the control group. As the diameter of bronchioles was less than 100μm and 100~200μm respectively, the average area of smooth muscle in bronchioles increased to 3.06-fold and 2.13-fold respectively. By electron microscopic, the proportion of Clara cells of respiratory bronchioles in COPD group decreased, the Clara cells were denaturalized, the proportion of Clara cells containing low density secretory granules increased. The airway resistance was 1.84 times while the dynamic compliance was only 70.45% comparing with the control group.
结果表明:(1)光镜下,COPD组大鼠与正常对照组比较,肺泡壁破坏、肺泡腔扩大,平均肺泡面积和平均肺泡直径分别为正常对照组的2.46倍和1.42倍,在直径<100μm及直径100~200μm的小支气管,COPD组大鼠支气管平滑肌面积分别为正常对照组的3.06倍和2.13倍;电镜下可见,在呼吸性细支气管,Clara细胞占上皮细胞的比例下降,细胞变性,含低电子密度颗粒的Clara细胞比例增加;呼吸功能测定可见COPD组大鼠呼气相气道阻力是正常对照组的1.84倍、动态呼吸系统总顺应性为正常对照组的70.45%。

In Part 1 of our research, BGSCs were sorted through immunomagnetic beads marking by CD133 and cultured in vitro, and character as a stem cell was identified by stem cell markers (CD133 and Nestin) and differentiated cell markers [ microtubule-associated protein 2(MAP2), glial acidic fibrillary protein and myelin basic protein] , ultrastructure observing with electron microscopeand engrafting to severe combined immunodeficiency mice for tumorigenesis test. The results were as following: Only a small subset of CD133+ glioma cells in glioma cell lines and fresh specimens from various pathologic grade could express stem cell markers CD133 and Nestin, view ultrastructure of a stem cell and be capacity of serial passage in culture. These CD133+ cells possese a marked capacity for multipotent differentiation and could differentiate into tumor cells expressing MAP2,β-TubulinⅢ, GFAP and MBP; When engrafted into SCID mice, they can generate and form tumors that phenotypically resembl the tumor from the patient.
在本课题中,在第一部分实验中采用以CD133为标志的免疫磁珠法从人脑胶质瘤组织和细胞株中分离脑胶质瘤干细胞并进行体外培养,通过免疫荧光技术检测干细胞标志物CD133、Nestin,诱导分化后检查分化细胞标志物MAP2、GFAP、MBP以及电镜超微结构观察和移植SCID鼠致瘤实验,对其干细胞特性加以鉴定,得到如下结果:不同病理分级的新鲜胶质瘤标本和胶质瘤细胞株中存在一小部分CD133+的胶质瘤细胞,能表达干细胞的标志物CD133和Nestin,符合干细胞的超微结构特点,体外培养能连续传代;具有多向分化潜能:诱导分化后能产生MAP2、β-TubulinⅢ、GFAP、MBP染色阳性的细胞;移植SCID鼠后能形成与亲本肿瘤表型一致的移植瘤。

Under the transmission election microscope, we had observed that the state of oocytes arid the cumulus cells around them was tightness, and there were plenty of long and thin microvilli playing the role of connection in the enwrapping. The shape of cumulus cells nucleus was ovate, and the longer and shorter diameters of the cumulus cells were 4.250±0.042μm and 2.750±0.07 μm respectively. The zona pellucida was well proportioned and tightly linking with oocyte. There were great deals of multi-crista mitochondria, lipid droplet and vacuolus. The mitochondria longer and shorter diameters were 0.600±0.106μm and 0.490±0.117μm. Granular nucleoplasma distributed very well in the cell nucleus.

For even more adapting net load, it proposes a control strategy by weight and adaptive adjustment to ensure XML streams quality.
