According to the characteristic of silver nanoparticles, their uniform dispersion in reaction media was obtained by surface modification, ultrasonic radiation and the select of emulsion, and then emulsion polymerization system were formed by combination with reactive monomer and initiator. Nano-Ag/Polystyrene core-shell particles were prepared by emulsion polymerization and their microstructure were characterized by TEM, FTIR, XPS, AFM and TG/DTA. The experimental results showed that silver nanoparticles were coated by polymer and the composite particles had typical core/shell structure, and the microlevel combination was realized. Meantime, the form mechanism of nano-Ag/polystyrene core-shell particles was tentatively researched. The conclusion is that the perfect coating was able to obtained by the control of reaction condition.
根据纳米银粒子的特性,借助于表面处理、超声辐照以及乳化剂的选择,使其均匀分散在反应介质中,再与反应单体、引发剂等形成乳液聚合体系;采用乳液聚合方法、包裹聚合方式,制备出纳米银/聚苯乙烯核壳复合粒子,并借助于TEM、FTIR、 XPS、AFM、TG/DTA等分析手段对复合粒子微观结构进行了表征;结果表明:纳米银粒子表面被聚合物层包覆,复合粒子呈明显的核壳结构,实现了微观尺度上的复合;同时,初步探讨了纳米银/聚苯乙烯核壳复合粒子的形成机理,指出:通过控制反应条件,可以使形成纳米银/聚苯乙烯核壳复合粒子的聚合反应方式占主导地位,实现最大限度地包覆。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.
研究方法:(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC,细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构;(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定;(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC,并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况,根据各个细胞克隆受Dox调控表达的程度,选择低背景、高诱导表达AT2R的细胞系,作为双重稳定MSC细胞系;蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况;(4)建立大鼠颈动脉球囊损伤动物模型,将双重稳定MSC在术中导入血管,分别于14 d、28 d进行病理切片,检测可调控AT2R对新生内膜增生的影响;采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变,TUNEL法检测血管组织中细胞凋亡的变化情况。

The mian works are as follows:(1)By coprecipitation technique and coprecipitation- gel technique the nano zirconia powder was produced, factors affecting the properties of nano zirconia powder such as pH and the content of stabilizator are analyzed;(2)By the new pressless sintering process, the nano zirconia ceramic was produced, factors which can affecting the properties of nano zirconia sinter body such as moulding press and the sinter temperature are analyzed;(3)By the microtherm pressless sintering process, the nano copper added functional zirconia ceramic composite was produced, microscopic dimensionality and electric properties of such sinter body and the factors affecting such sinter body are analyzed;(4)By the self-invented "external albumen coating" technique the zirconia powder was successfully coated by carbon in the corresponding sinter body, which can help solve the traditional aggregation problem to some degree;(5)Based on analyze the characteristics in the nano powder sinter process, the "three-ball sinter model" was proposed, for the relationship between the relative density and porosity of sinter body, by mathematical fitting, such model was successfully verified.

Chicken IL-15(ChIL-15), Which was discovered in 2001, plays a main role in activating NK cells and CD8+ memory T cells, and may therefore have potential as a vaccine adjuvant.According to published ChIL-15 gene sequence, a pair of primers were designed and synthesized to clone ChIL-15 cDNA from ConA-activated chicken splenocytes by RT-PCR. Recombinant plasmid pUC19ChIL-15 carrying the ChIL-15 gene was constructed. The gene encoding mature ChIL-15 (mChIL-15) was amplified from pUC19ChIL-15 by PCR and cloned into pMD 18-T vector. After digested with Kpn I /Pst I , the mChIL-15 gene was subcloned into prokaryotic expression vector pPRoEX橦Ta and transformed into E.coli DH5a .The transformant was induced by IPTG, and the recombinant ChIL-15(rChIL-15) was expressed as a fusion protein with a histidine hexamer tag at the N-terminal end of the protein. Following expression, the protein was purified by ProBond?Nickel-Chelating Resin. The uninduced and induced protein lysates and the purified protein were separated by SDS-PAGE and transferred onto nitrocellulose membrane to identify rChIL-15 by Western blot. The rChIL-15 was used to immune the guinea pig to obtain rChIL-15 polyclonal antibody.
