In this paper we combined three chromatographic separation and purification technique such as affinity chromatography, ion exchanger chromatography and hydrophobic interaction chromatography to develope a new technology of stimutaneous extraction of three enzyme from pancreatin. The different fishes which are employed in the present studies are wild-type salmon, cultured salmon of freshwater and seawater, sea perch and fat greenling. The complete CT gene sequences of salmons are obtained by PCR amplification. The partial CT sequences of sea perch and fat greenling are obtained by in vitro cloning PCR method. Alignment of obtained CT sequences with other fish CT shows that CT appears to be well conserved among the same family. And the relative in taxonomy is far away, the similarity of different fish CTs is low. On the contrary, the closer the relative in taxonomy is, the higher the similarity of different fish CTs is. The sCT is expressed in pGEX-4T-X by recombinant form . We also succeed in the research of sCT expression alone by expression PCR. In addition, sCT antiserum is obtained using GST-sCT as antigen, and the high titer is tested by double immunodiffusion. In the rat bioassay, administration of 50 μg recombinant protein evoked significant hypocalcemia at 1 h after the data are analyzed by t-test.
本文用PCR方法克隆了野生鲑鱼、养殖鲑鱼的降钙素基因,并应用体外克隆PCR的方法首次克隆出鲈鱼、六线鱼降钙素的部分基因序列,通过对克隆的降钙素序列的比较研究,结果显示同一科的鱼降钙素序列保守性较高,同时,根据降钙素的部分氨基酸序列进行了降钙素相似性的比较研究,结果显示在分类学上,分类地位较远的鱼,其降钙素相似性较低,分类地位越接近的鱼,其降钙素相似性越高;我们利用谷胱甘肽S-转移酶(Glutathions S-transferase,GST)融合表达载体pGEX-4T-X对克隆的鲑鱼降钙素基因进行了融合表达研究,应用表达PCR的方法对降钙素基因的独立表达进行了初步的研究探索,并将纯化的融合蛋白作为抗原,获得了高效价的兔抗鲑鱼降钙素免疫血清;生物活性研究表明,大肠杆菌表达的融合蛋白具有显著的降血钙作用。

According to the studied results at present, the studied objects are mainly thin plate and beam about sound radiation mode, a method computed sound radiation mode of complex structure is presented in this dissertation, according to the results of the previous two chapter, a theoretical method to solve acoustical radiation mode and acoustical radiation efficiency of complex structure by boundary element method and acoustical radiation theory is posed, the acoustical radiation power is expressed as a Hermitian quadratic form, while the radiation modes is determined by general eigenvalue decomposing, and then the radiation efficiency is computed via the orthodoxy of radiation modes with regard to impedance matrix and average velocity matrix; lastly, the validity of the method is proved by pulsating sphere and radiating cube with analytical results. According to results of the form three chapter, the structure sound radiation sensitivity is studied in detail, the sound radiation power of structure can be expressed as positive Hermitian quadratic form, the sound radiation sensitivity can be expressed as two parts by partial differential with respect to design variable, which are sensitivity of boundary velocity and impedance matrix, combined with the theory of FEM and BEM, the structure sound radiation can be translated to the analysis of structure dynamic sensitivity and impedance matrix sensitivity. The theory posted in this dissertation is tested by the FEM software ANSYS and the software AAS programmed by author.

The conversion boundary of self-incompatibility was researched by sowing by stage. Sunshine of day and temperature was studied critically by investigating leafage and researching traits indoors. The results showed that the daily minimum temperature was the principal factor of affinity conversion and the affinity conversion boundary was 16.5~18.1℃.HE97 was primarily regard as self-incompatible line in this research. The method was by inbreeding and sib HE97, Half the silk of HE97 and Zheng58. Inbreeding and cross have been done simultaneously. The extending condition of pollen tube was observed by dying the silk after pollinating 4 hours. The individuals with pollen tube were observed having seeds, while the individuals without pollen tube were observed having few seeds or no seeds. So this study regard HE97 as sporophytic self-incompatibility .The planting ration of female parent HE97 and male parent 87-1 was 4:1.The nature cross-pollination was studied by the theory of endosperm xenity, the study show that the setting percentage of HE97 was 625%, in which the seed in self was 70% and the seed in hybrid was 30%. This was consistent with the study of silk anatomy structure, then come to the same conclusion: compatible pollen germination and fertilization can stimulate the self-incompatible pollen germination and fertilization in the same corn cob.
