Methods Monoclonal antibody against the glycophorin A was used to identify nucleated erythrocytes in the peripheral blood of pregnant women.Candidates fetal cells from 3 pregnancies at 16 to 24 weeks of gestation were isolated successfully.Sorted cells were also confirmed using Y-special probe (PY3.4) repeated sequences in maternal blood from women carrying male fetuses were detected by polymerase chain reaction.Results The PY3.4 position hybridization rates were 31.5%0 and 38% respectively in sorted GPA positive cells of 3 samples by FISHand the results of GPA+ cells of the 3 cases by PCR were positivenegative and positive respectivelywhich were manifested by the gender after aborticide.

The results showed:(1) the cellule morphous and the disposition in chick embryo of PGCs were coincident no matter stained by PAS or HE staining, and HE staining could disclose the morphologic characteristics more clearly, exactly and completely;(2) genesis of blood island could be observed at the boundary of light and dark region of the extraembryonic blastoderm at about 26 hours;(3) both the blood vessel endothelium cells and free cells of the blood island were differentiated from PGCs. The generating of genuine yolk sac was at about 44-48 hours.

Results: The results showed that:(1) The three types of NOS immunoreactivity were not found in rat testis until 30 days after birth;(2) NOS1 immunoreactivity was found in a few spermatocytes in rat testes on postnatal 30 days, and in spermatozoa present at the lumen surface of seminiferous tubules and some Leydig cells on postnatal 60 days;(3) NOS2 immunoreactivity appeared in a few spermatocytes, Sertoli cells and peritubular myoid cells on postnatal 30 days. On postnatal 60 days, NOS2 immunoreactivity appeared in some Leydig cells, peritubular myoid cells, Sertoli cells, very few spermatocytes and the head of immature spermatozoa in some seminiferous tubules;(4) NOS3 immunoreactivity was detected in a few spermatocytes on postnatal 30 days, as well as in the smooth muscle cells of blood vessels on postnatal 30 days and 60 days.
