Results Same conservative sites and key catalytic sites existed among SjLDH and LDHs from other species. Similarity of SjLDH compared to CsLDH, TvLDH and HsLDH was 75%, 17%, 58%-60% respectively. Phylogenetic analysis demonstrated that the evolution relation between SjLDH and DmLDH was closer than the relation between SjLDH and CeLDH, the relationship between SjLDH and HsLDH-B,-C was closer than HsLDH-A. Three trans-membrane regions were found, the region of 98aa-106aa in three hydrophilia regions located outside of membrane was inferred as the major antigen epitope. This antigen epitope had significant difference with LDHs from protozoon ( Pf ., Tg ., Tv .) and had 1-3 amino acid residue difference with MmLDH, HsLDH-A,-B,-C, and was the same with CsLDH. One of the key catalytic residues and substrate binding loop were located in this region. Tertiary structure demonstrated that 98aa-106aa was on the surface of the protein and formed a substrate binding loop, other two key catalytic sites were at the position near the loop.

It could be deduced that all the insecticides tested in this experiment damaged the spiders′ intestinal mucosa, compromising its function as a barrier and letting large molecules such as FTIC-D more easily penetrate. Among the insecticides, pyrethrin and Bt preparation are bio-insecticides, and the FITC-D concentrations of the spiders exposed to these two bio-insecticides were more than those of the spiders exposed to the chemical insecticides imidacloprid and dipterex. The difference between the bio-insecticide groups and the imidacloprid group was significant, but there was no significant difference between the bio-insecticide groups and the dipterex group.

Fluorescence resonance energy transfer happens from CdS to Rhodamine B, Butyl rhodamine B and Rhodamine 6G, respectively. There is a higher FRET efficiency between CdS and Neutral red with the value being 0.282. Moreover, coupling reaction between -NH_2 of Neutral red and -COOH of CdS -TGA leads to red shift of CdS fluorescence spectrum. The reaction also occurs between Safranine T and CdS. At the same time, the Fluorescence of ST is quenched by forming ST -CdS ground state complex. Chapter 6, CdS QDs modified by chitosan and polyethyleneimine can be quenched by Cu~(2+). According to the experimental result, we develop the novel sensor for Cu~(2+).

During the four years study on the project, we develop a dynamical forecast model that describing the subtropical ridge surface. A new theory on the role of the land-sea thermal difference on the formation of the Subtropical High is advanced. It is concluded that in the summer subtropics over each continent and its adjacent ocean LO, SE, CO, and a double-dominant heating from west to east compose a LOSECOD heating quadruplet. The influence of the Eurasia continent on SH varies according to the seasonal variation. From the ideal numerical simulation it is shown the important role of the Eaiuan continent and the huge topography on the mechanism of formation and mantanence of the SH during the summer. It is verified by the numerical simulation that the extension of the subtropical continent into the tropics greatly enhances the " East Asian monsoon". Through the Rossby energy dissipation theory it is verified that the deep latent convective heating induces the onset of the South China Sea monsoon over the Bay of Bengal. The relationship and mechanism between the precipitaiton over the East Asia and the Subtropicla high is illustrated by the data. It is also found the close phase lock between the east-west shift of the SH and the disturbence of the east wind belt over the tropical troposphere. The interaction between the structure and activity feature of the SH and the Meiyu over the Yangtze-Huaihe river valley is revealed from the data. The numerical simulation verified the impact of the different SH pattern on the path of the typhoon.
