ABSTRACT Objective: To observe the protective effects of Corydalis ambailis migo total alkaloids on cerebral ischemia/reperfusion injury in rats and to investigate its mechanism.

摘要］ 目的：观察夏天无总碱（Corydalis ambailis migo total alkaloids， COAMTA）对脑缺血/再灌注损伤的保护作用并对其机制作初步探讨。

The results showed as follows: 1 Extraction of gelsemium total alkaloids. Total alkaloids were extracted with 95 per cent ethanol. The extract rate was 0.519 per cent and the extract purity were 88.02 per cent and 87.38 per cent; Total alkaloids were extracted with chloroform .The extract rate was 0.416 per cent and the extract purity were 89.02 per cent and 94.52 per cent. 2 Isolation of gelsemium total alkaloids with TLC. Total alkaloids contained five or more alkaloids. The optimal developing agent was chloroform :methanol :ethyl acetate(9:l:0.3),the optimal eluent was methylene chloride:methanol(92:8). 3 Isolation of total alkaloids with column chromatography. Alkaloid A and alkaloid B were obtained, alkaloid B was identified as koumine.

研究结果如下：1、钩吻总碱的提取：用95％的乙醇提取钩吻生物碱提取率为0.519％，其提取物纯度为88.02％、87.35％；用氯仿提取钩吻生物碱提取率为0.416％，其提取物纯度为89.02％、94.52％。2、钩吻总碱的薄层层析分离：总碱中含有5种或5种已上的生物碱；氯仿：甲醇：乙酸乙酯(9：1：0.3)是最佳薄层层析的展开剂；二氯甲烷：甲醇(92：8)是柱层析的最佳洗脱剂。3、钩吻总碱的柱层析分离：共得到两种钩吻生物碱单体成分碱A和碱B，碱B为钩吻素子，碱A可能是钩吻素甲。4、钩吻总碱的急性毒性试验：钩吻总碱小鼠腹腔注射的半数致死量LD_(50是：7.38mg／kg，95％的可信限范围为：6.67mg／kg～7.94mg／kg。

This thesis summarized the recent research of lycopodium alkaloids, including the structures of those new alkaloids, studies of the interaction between acetylcholinesterase and its inhibitors by NMR, and the synthesis of lycopodium alkaloids.

The chemical constituents of Huperzia serrata Trev. are mainly flavonoids, triterpenoids and alkaloids. In order to get further comprehension of lycopodium alkaloids, we studied the low-polar section of the alkaloids extraction of Huperzia serrata and 19 alkaloids were isolated by ways of silica gel, RP-18 and Sephadex LH-20 etc. All of their structures, including five new compounds and two known ones that were firstly found from this genus, were elucidated with the help of Mass, IR, NMR spectra and so on. The five new alkaloids (HS-07, HS-08, HS-09, HS-10 and HS-11) are belonged to lycopodine class. The other two ones that were new to this genus (HS-18 and HS-19) are belonged to miscellaneous group.

蛇足石杉的主要化学成分为黄酮，三萜和生物碱，为进一步了解该植物的生物碱成分，我们研究了蛇足石杉氯仿总碱低极性部位的生物碱成分，运用正相硅胶、反相RP-18、凝胶LH-20等方法分得了19个生物碱，并通过质谱、红外、核磁共振等波谱鉴定了它们的结构，其中5个为新化合物，2个为首次从该属植物中分离得到的化合物，所分得的新化合物：HS-07、HS-08、HS-09、HS-10和HS-11均为lycopodine类石松生物碱，首次从该属植物中分得的化合物为miscellaneous类：HS-18和HS-19。

The extraction and separation of total alkaloids from Wu-He Dipsacus by continuous extraction are described.Then the analysis of the quantity of alkaloids is carried out by UV method.Furthermore,the determination method of alkaloids is discussed.

Abstract］ Objective: Study and explore the express of gene and protein of protein kinase B about cerebral ischemia effects of phenolic alkaloids from Menispermum dauricumon cerebral ischemia in rats.

目的：探讨蝙蝠葛酚性碱（phenolic alkaloids from Menispermum dauricum，PAMD）预处理对脑缺血后大脑皮质细胞凋亡蛋白激酶B(protein kinase B，PKB)表达的影响。

METHOUS: An aqueous extract of Rhizoma menispermi, an alcohol purified extract named phenolic alkaloids of menispermum dauricum and Rhizoma menispermi polysaccharide were isolated and purified by using the three-step method.

用三步法初步分离、纯化北豆根提取物，得到北豆根水提液(aqueous extract of rhizome menispermi， RMAE)、醇提液的初步纯化产物蝙蝠葛酚性生物碱(phenolic alkaloids of menispermum dauricum， PAMI)和北豆根多糖成分(rhizome menispermi polysaccharide， RMP)。

The results showed: when TLC solvent system was: chloroform - methanol - diethyl amine = 90:9:1, alkaloids from lotus leaves were well isolated, and five components were isolated and identified by spraying Dragendorff reagent. The optimal conditions of HPLC solvent system were: methanol- water - diethyl amine = 75:25:0.0125. Under the conditions, components of alkaloids from lotus leaves were separated very well, and ideal RP-HPLC peaks were obtained.5、Methods of High Speed Counter Current Chromatography and preparative High Performance Liquid Chromatography applied to identify and purify alkaloids from lotus leaves were set up. When solvent system of HSCCC was: chloroform - methanol - water (pH=4.00)= 4:3:2, speed of chromatogram was 700rpm, flow speed of the mobile phase was 2mL/min, four pure components relatively were attained by isolation and purification.
