Methods: Eleven B-CLL patients were studied. Leukemic lymphocytes with (n=8) or without (n=3) P2z receptors were exposed in vitro to ATP, benzoylbenzoic-ATP, 2-methylthio-ATP(2MeSATP), adenosine-5′-［γ-thio］ triphosphate, and other nucleosides for 8h. Apoptosis was detected by electron microscopy, agarose gel electrophoresis, and quantitative TdT assay. Results:Apoptosis was detected only in leukemic lymphocytes with P2z receptors. By using the quantitative assay, ATP-inducing DNA strand breaks were found to occur specifically for BzATP, ATP and 2MeSATP, but not for ATP-γS and other nucleosides.

The thymuses, spleens, sera and anticoagulant blood were collected after the rats were killed, to carry out these works:(1) Zinc concentrations were analyzed by atomic absorption spectrophotometry;(2) Body weight and the weight of thymuses and spleens were measured, the organ index were calculated;(3) Changes in pathology of thymus and spleen were observed;(4) Using TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling) method, the apoptosis of thmocytes and spleen lymphocytes was checked;(5) The expression of bcl-2、bax mRNA in thymus and spleen were detected by RT-PCR (reverse transcription polymerase chain reaction);(6) The expression of p56〓, a signal transduction protein in thymus and spleen were detected by immunohistochemistry;(7) Two-color cytofluorometric analysis was used to assess the expression of CD4, CD8, CD45RA and CD45RC in peripheral blood lymphocytes.

The test groups were drenched orally with different dosages of compound Radix scutellariae granules. After being drenched orally for ten days, their thymus-index and spleen-nidex, immune globulins of IgA, IgG, IgM and alexins of C3, C4 of serum were defermined, and experiments in vivo were done. The mice-spleen's lymphocytes which were cultured in vivo were added with different concentrations of compound Radix scutellariae granules. The lymphocytes translation stimulate indexes were measured. The ability of licking up neutral red of the peritoneal macrophages cultured in vivo was measured in the groups.
