The gonadotropin releasing hormone receptor gene was studied as a candidate gene for the prolificacy of Small Tail Han sheep. Eight pairs of primers were designed to detect single nucleotide polymorphisms of exon 1, exon 2 and exon 3 of GnRHR gene in both high fecundity breeds and low fecundity breeds (South African Mutton Merino, Corriedale and Chinese Merino sheep) by PCR-SSCP. Only the products amplified by primers P4 and P7 displayed polymorphisms.

The inhibin a gene、 the bone morphogenetic protein 4 (BMP4) gene and the bone morphogenetic protein 7 (BMP7) gene were studied as candidate genes on the prolificacy of Small Tail Han sheep. Single nucleotide polymorphisms of 5 ' regulatory region、 exon 1 of INHA gene;exon 2、 exon 3 and exon 4 of BMP4 gene and exon 2、 exon 3 of BMP7 were detected in high fecundity breed and low fecundity breeds (Chinese Merino sheep, Corriedale sheep and South African Mutton Merino sheep) by PCR-SSCP. The results indicated that there was a mutation (316C→T) in 5' region of INHA gene in Small Tail Han sheep, Chinese Merino sheep and Corriedale sheep, the same mutation did not exist in South African Mutton Merino sheep.

Polymorphism was detected for exon 3 of BMP4 gene in four sheep breeds. For exon 3, three genotypes were detected in four sheep breeds. In Small Tail Han, Chinese Merino, Corriedale and South African Mutton Merino sheep, frequency of genotype AA was 0.017, 0.216, 0.115, 0.429, frequency of genotype AB was 0.102, 0.317, 0.269, 0.500, frequency of genotype BB was 0.881, 0.467, 0.616, 0.071, respectively. Sequencing revealed one singlenucleotide mutation C→A at 305 bp of exon 3 of BMP4 gene in genotype BB in comparison to genotype AA, and this mutation resulted in an amino acid change of alanine→aspartic acid.

Segments of Adh gene in azuki bean (variety KS 5), covering an interval between partial intron 3 and intron 9, were amplified and sequenced with two pairs of primers. The result has proven the viability of using conserved sequences in a completely sequenced gene of one species to clone same gene in another species. In the comparison between Adh genes of azuki bean and pea, high sequence similarity (about 80%) in exon and long length difference (up to 116 bp) in intron were found. Besides, the edge regions of Adh introns with GT/AG counterparts and more conserved sequences than central regions was observed in both species.

The result of uroscopy of the proband was strong positive. There was a novel deletion mutation of c.876-877 del TC in the coding region of exon 6 of IDS gene, which was a hemizygous mutation. However, the mutation of his mother and sister was a heterozygous mutation. Detection of the exon 6 of IDS gene showed that the mutation was not found among normal controls and other patients with MPS I, IV, and VI other than MPS II. Homology comparison of amino acid sequences from different species showed that the phenylalanine glutamine of the mutation site of c.876-877 del TC located in p.292-293 was highly conserved. The activity of IDS enzyme of the proband was only 2.3 nmol/4 h/mL, which was much lower than normal; but the activity of IDS enzyme of his father, mother and sister was 641.9 nmol/4 h/mL, 95.8 nmol/4h/mL and 103.2 nmol/4h/mL, respectively.

Methods The 26th exon, the 30th exon and the 21st intron of gene GP Ⅱb in 110 individuals were amplified by polymorphism and sequenced to investigate whether there was linkage among the polymorphisms of the gene. Human platelet antigen-3 (HPA-3) gene frequency was detected by Fok 1 enzyme in 147 patients with hematologic diseases, and was compared with that in 110 normal individuals. Forty-four patients who received apheresis platelet transfusion repeatedly were randomly divided into the HPA-3 homotype group and the control group. The antibodies of the platelet were detected after 3 times of platelet transfusion.
