Moreover, there apparently existed morphologic changes in this course, and there maybe existed the apoptosis, which correlated with bacteria propagation and acid producing. With the culture time postponed, the nutritive substances in the culture medium lacked also, and the morphologic change and apoptosis appeared more obviously.

Mixed saliva and pooled plaque samples from the smooth surfaces of teeth were collected for microbiologic analysis before administration (baseline~ day 0).. during treatment (days 7.. 14 and 21) and after withdrawal of the agents (days 28~ 42 and 56) and plaque index were recorded. Microbial cultivation were performed by serially diluting the samples with PBS solution and cultivating the aiquots on Mitis Salivarius Agar and MS supplemented with bacitracin for Oral streptococci and Mutans streptococci respectively. The plates were incubated at 37~C under microaerophilic condition(90%N2,10%CO.,) for 48 hot:rs. The number of colony-forming unit per plate was counted and the percentage of Mutans Streptococci per Oral streptococci in dental plaque were determined.

The PCR based assay was tested on 19 strains isolated in throats from patients with guttate psoriasis. The results demonstrated that 6 strains were positive in 15 streptococus strains among which there were 3 streptococus pyogenes, 1.S.agalactiae, 1 S. eguisimilis and 1 S. uberis, and 4 non- streptococus strains were negative. Emm6 was found not only in suppurative hemolytic streptococus, but also in S.uberis.

A series PCR amplification for differential control strains and DNA samples diluted gradient (1:10) have been used to evaluate the specificity and sensitivity of PCR assay established.Results 1. Detection of GAS by PCR assay: The 345bp specific fragment of speB gene were amplified in all the tested GAS strains including three strains of scarlet fever, whereas it was detected in none of the differential control strains. The lowest limit of detection was 6.5pg genome DNA of GAS strain. 2. Detection of corynebacterium diphtheria by PCR assay: The318bp specific fragment of toxB gene were amplified in all the tested toxigenic corynebacterium diphtheria strains, whereas it was detected in none of the differential control strains. The lowest limit of detection is 850fg/μl genome DNA of corynebacterium diphtheria strain. 3. Detection of Lp by PCR assay: The 340bp specific fragments of mip gene were amplified in all the tested Lp strains, whereas it was detected in none of the differential control strains including three strains of non-pneumophila.

Using this model three competence-deficient pneumococci, which was identified by PCR and transformation experiment, were constructed. It is found different strain has different bacterial optimal density for being induced into competence. The optimal density for strains 1, 2 and 22 was 0.1, 0.08and 0.07 respectively. The optimal condition for transformation is c plus y culture at pH8.0. In c plus y culture at pH6.8, which restrained pneumococci forming transformation naturally, pneumococci could be induced to form transformation by CSP.It is concluded that we could induce different S.pneumoniae by CSP into competence in their optimal density in laboratory. 2. It was discussed the relationship between pneumococcal transformation formation and its opportunistic pathogenesis. Comparing their virulence with that of parental strains by challenging mice intraperitoneally and adhering to ECV-304 cell respectively.
