On the basis of the kinetic equation of substrate reaction in the presence of urea or guanidine hydrochloride, all microscopic kinetic constants for the free enzyme and enzyme-substrate binary and ternary complexes have been determined. The results of the present studies indicate that:①In the presence of urea or guanidine hydrochloride, enzyme-substrate complexes lose their activity less rapidly than the free enzyme. Therefore, both substrates, NADPH and 7, 8-dihydrofolate, protect dihydrofolate reductase against inactivation.②The denaturation of dihydrofolate reductase by urea follows single-phase kinetics, and changes in enzyme activity and tertiary structure proceed simultaneously in the unfolding process, so it may be an"all or none"process.③The GdnHCl-induced unfolding of the dihydrofolate shows a biphasic transition, while the change in the enzyme activity is a single exponential process. The rate constant of inactivation is consistent with that of the fast conformational change. Therefore, the kinetic intermediate of protein unfolding should be a partially folded and inactive form.

There is still no consensus on the expression of the two isoforms in the human prostate and their cellular locations are also controversial. It is not known to which extent each of these isozymes contributes to overall enzyme activity in different prostatic tissues. In an attempt to elucidate which isoform of the 5α-reductases is responsible for the metabolism of T in normal and diseased human prostate, we performed qualitative and semi-quantitative studies of both enzymes in normal, benign hyperplastic and adenocarcinomatous human prostate tissues by RT-PCR and Immunohistochemistry methods.

The result shows: in 0~360 kg/hm^2 fertilizer can improve the plant height, dry matter accumulation of maize, nitrogen accumulation, root vigor, leaf nitrate reeducates activity, chlorophyll content and yield, meanwhile the leaves' nitrate reeducates activity, dry matter accumulation and the final output of wheat, root activity and other indicators were significantly correlated; Cultivating under the same conditions and dropping the fertilizer amount from 360 kg/hm^2 to 315 kg/hm^2, the dry matter accumulation will be increased, the roots vigor and leaf nitrate reeducates activity will be strengthened, but the yield are not obviously different in the same treatment.
