After treatment with 50 mmol·L-1 crocin, the inhibited growth of tumor was observed. Electronic microscope was used to observe the morphological changes. The expressions of Bcl-2, Bax, Survivin and Cyclin D1 were detected by immunohistochemistry.

Results:①The amount of human colon carcinoma cell line SW480 treated by quercetin decreased. The morphology of partial SW480 cells was shrunk volume, integrated cell membrane, condensed cytoplasm, pyknotic chromatin, nuclear fragmentation. Apoptotic Corpuscles were found by electron microscope.②MTT colorimetric assay showed quercetin inhibited the growth of human colon carcinoma cell line SW480 in a time- and dose-dependent manner when the concentration of quercetin was 30、60、90μmol/L.③Flow cytometry analysis showed the cell cycle of SW480 cell was restricted in G1/S. G0/G1 phase rate increased and S phase rate decreased with increasing concentration of quercetin and time lasting.④ Zymogram analysis assay showed the secretion of matrix metalloproteinases in human colon carcinoma cell line SW480 treated by quercetin decreased. With increasing concentration of quercetin, the secretion of MMP-2 and MMP-9 decreased.⑤Immunohistochemistry method demonstrated the position expression of Cathepsin-D in SW480 cell was suppressed by quercetin in a time- and dose-dependent manner.
研究结果:经槲皮素处理的人结肠癌SW480细胞数量减少,部分细胞体积缩小,细胞膜完整,胞浆浓缩,核染色质固缩,细胞核碎裂,形成凋亡小体;MTT法检测显示当作用浓度为30μmol/L~90μmol/L时,槲皮素对人结肠癌SW480细胞的生长有抑制作用,其抑制作用随着作用浓度的增加和作用时间的延长而增强;流式细胞学发现槲皮素主要作用于人结肠癌SW480细胞周期的G1/S期,大部分细胞被阻断于S期,随药物浓度的升高和作用时间的延长,G0/G1期细胞比例逐渐增加,S期细胞比例逐渐减少;酶谱分析法检测显示不同浓度的槲皮素能够抑制人结肠癌SW480细胞分泌MMP-2及MMP-9,随浓度的升高,MMP-2及MMP-9的分泌量减少;免疫组织化学法显示不同浓度的槲皮素处理人结肠癌SW480细胞后,Cathepsin-D的表达随药物浓度的升高和作用时间的延长而降低。

The thesis thinks the present conditions and characteristics of appeals for help involved in lawsuit are that appellants are nature persons primarily, appealing causes are to refuse to accept decisions and reflect to execute improperly, cases type are civil cases primarily, as well as showing those characteristics that appeal case are getting long-term, group appeal are getting organizing and appeal action are getting non-rationalizing and so on from the angle of a judge who is engaged in the administration of justice, and on the basis of relevant data which are a case of appeals for help involved in lawsuit of two level courts and centralized dealing ways of autonomous prefectures.

Through analysis and discussion in the following chapters, four strategies are summarized for teachers' further reference.(1) The individualized physical education program-Through the plan with similar to IEP, teachers arrange physical activities properly for children with mental retardation.(2) Cognitive behavior modification-Through demonstration and guidance, students are able to change behaviors by modeling and correcting their cognitive patterns.(3) Task analysis approach-Teachers have to divide a particular movement skill into several individual sub-skills and then students would be able to master that specific skill by linking every well-practiced sub-skill.(4) Data processing mode-Students are motivated to learn with fun because the target skill has been analyzed into meaningful and organized procedures.
经过分析与探讨,本文提供四大体育教学策略供从事体育教学之教师参考:一、个别化体育计划——透过类似个别化教育计划(Individual Education Program, IEP)的计划,让教师先对智能障碍学童的需求与体育活动作适当的安排;二、认知行为改变技术——教师以示范、引导的方式,使智能障碍学童经由模仿,修正认知,让行为得以改变;三、工作分析法——教师先将动作技能分解成数个分项,智能障碍学童经充份练习每一分项,最后再将每个细部动作串联;四、讯息处理模式——透过教师引发学习动机,并将动作以有意义化与组织化的学习方式,让智能障碍学童从乐趣中学得动作技能。
