The RAPD was used to study the genetic diversity of 19 KinXin series potato cultivars. The result showed that 52 RAPD bands were amplified using 7 primer, among them 43 bands were polymorphic, and 4~9 bands were amplified in each lane. The RAPD analysis indicated that the genetic distance of 19 potato cultivars was between 0.17~0.72, average value was 0.39. When all of the polymorphic bands were analysed by cluster analysis, 19 potato cultivars were classified into 3 groups. The analysis showed that there existed close sibship among the majority of potato cultivars, and the genetic distance between the minority cultivars and the others is longer, which indicate that the genetic basis of KeXin series potato cultivars is wide.
利用RAPD标记技术对19份克新系列马铃薯品种进行了遗传多样性分析,分别提取19份马铃薯品种的DNA,进行随机引物多态性扩增,从1000条随机引物中初步筛选出7条有多态性的引物进行详细研究,每条RAPD引物扩增出4~9条带,共获得52条带,其中多态性条带为43条;19份马铃薯品种的遗传距离介于0.17~0.72之间,平均值为0.39,平均遗传距离介于0.31~0.51之间;聚类分析结果在GS=0.53处可将克新系列马铃薯品种划分为三类,聚类结果与系谱分析基本相符,同时也说明克新系列马铃薯的遗传基础有所拓宽。

Genetic diversity among 7 populations of Trillium tschonoskii were analyzed by using inter SSR and amplified fragment length polymorphism molecular markers. The results were as follows:(1)ISSR: 12 out of the 100 primers showed consistent banding patterns and amplification, a total of 135 bands were produced with an arrange of 250-2500bp, 46 of which (34.07%) were polymorphic.

The average percentage of polymorphic bands are 72.06% for ISSR and 66.67% for RAPD respectively. The genetic diversity of ginkgo revealed by ISSR (the average effective number of alleles is 1.8780, the average Nei"s gene diversity is 0.4652, and the average Shannon"s information index is 0.6574) are all higher than those by RAPD (the average effective number of alleles is 1.7842, the average Nei"s gene diversity is 0.4317, and the average Shannon"s information index is 0.6211), while the standard errors of the parameters estimated by ISSR were lower. Therefore, ISSR molecular markers are more suited than RAPD molecular markers when testing the genetic diversity of ginkgo populations and determining the genetic relationship of among populations or among individuals which are much similar hereditarily.
本文应用RAPD和ISSR两种分子标记技术,选取中国5个可能的野生银杏居群共计75个样品,对其遗传多样性进行了研究,得出以下结论:(1)用筛选出的12个RAPD引物和10个ISSR引物进行PCR扩增,分别扩增出65条和68条重复性高、清晰的条带,多态性位点百分率分别66.67%和72.06%,ISSR揭示的银杏遗传多样性(平均有效等位基因数目为1.8780,平均基因多样度为0.4652,平均信息指数为0.6574)高于RAPD所得到的结果(平均有效等位基因数目为1.7842,平均基因多样度为0.4317,平均信息指数为0.6211),其所估算参数的标准差要低于RAPD所估算出的值,因此,在研究亲缘关系非常近的银杏物种的遗传多样性并试图确定居群间或个体间的遗传关系时,ISSR分子标记技术比RAPD分子标记技术更为合适。
