Methods Male infertility couples from October 2006 to July 2008 were chosen and divided into ICSI group and TESA+ICSI group. ICSI group were 584 oligozoospermia cases with alive normal sperm in spermatic fluid. TESA+ICSI group were 54 obstructive azoospermia cases with alive normal sperm in testicle. Two groups were treated with ICSI after wives obtained oocytes by ovarian stimulation.

We biopsied 1-2 single blastomere from 6-8 cell cleavage stage intracytoplasmic sperm injection embryo, and using nested PCR amplified the high frequently mutation region G380R of FGFR3 gene of the single blastomere. The products of PCR were digested by restriction enzyme Bfm Ⅰ, then the digested products were detected by 10% PAGE to see whether the embryo inherted the mutation of the patient and to screen out normal embryo transfer.

The results showed that after 2 h of culture in MEM the chromosomes of oocyte were seperated to form telophase I while a small spindle was observed around chromosomes of primary spermatocyte. However, two clear spindles were observed in the oocytes cultured in CB containing MEM. After further culture, the chromosomes of both primary spermatocyte and oocyte intermingled and formed one large spindle.
