In this study, Mono-nuclear cells containing hematopoietic stem/progenitor cells were isolated from umbilical cord blood of normal full-term deliveries, healthy adult and heterocellular HPFH patient bone marrow by density gradient centrifugation. After simple purification and expansion in vitro, MNCs were induced to erythroid cell differentiation using different culture systems and cytokines including Epo, IL-3 and GM-CSF.

RESULTS:Isoalantolactone exhibited excellent anti-proliferative activities on HeLa, SHIN3 and HOC-21 and HAC-2 cell lines, while 1-O-acetylbritannilactone and britannilactone demonstrated weak anti-proliferative activities on HeLa, SHIN3, HOAC-21 and HAC-2 cell lines even at the concentration of 100 μmol/L. The apoptotic rate in isoalantolactone group with the concentration of 12.5 μmol/L was higher than that in PBS group.

Light microscope and transmission electron microscopy showed that SMMC-7721 cells induced by SAHA had undergone the restorational alteration in morphology and ultrastructure, which were different from those of nontreated cells but were similar to those of normal cells, and the changes were as follows: the cells turned to be flat and spread; the nucleo-cytoplasmic ratio lessened and nuclear shape became rather regular; the number of nucleolus reduced and its volume lessened; euchromatin increased while heterochromatin decreased in nucleus; in the cytoplasm, mitochondria grew in number with relatively consistent structure and well-developed mitochondria cristae; Golgi complex turned to be well-developed and typical; rough endoplasmic reticulum increased. Immunocytochemistry assay showed that the expression of AFP and PCNA were declined significantly. FCM analysis showed SAHA could arrest SMMC-7721 cells in G0/G1 phase, with an accumulation of the cells in G0/G1 phase while a decrease of cells in S phase. Semi-quantitative RT-PCR detection revealed that the expression of p21WAFl mRNA was upregulated remarkably in the cells treated with SAHA.

To determine the effects of ovarian steroid hormones and some cytokines on the expression of RANTES in cultured cells METHODS AND RESULTS The study included the following two parts 1 The Protein Production of RANTES in Peritoneal Fluids and the Expression of RANTES mRNA in Eutopic and Ectopic Endometrium of Patients with Endometriosis Twenty-eight patients with endometriosis were selected as study group and twenty patients without endometriosis and maglinant disease were selected as control group . In our study, 10 samples of peritoneal fluid in Em and 10 samples of in En were taken, 13 specimens of ovarian endometrioma , 3 red peritoneal endometriotic lesions , 2 uterosacral ligament nodules , 17 matched eutopic endometrium and 12 specimens of endometrium in En were biopsied.

Results: Compared with control group, apoptosis cells increased from 0.5%to 10%(some even to 15%) after 24,48 and 72 h action of -300,-500 and -1 000 V electrets. After action of -500 V PTFE electrets for 48-72 h, fibroblast cells showed characteristic morphological features of apoptosis. These features included chromatin aggregation, nuclear and cytoplasmic condensation and partition of cytoplasm and nucleus into membrane bound-vesicles.
